Toward understanding recurrent meningioma: the potential role of lysosomal cysteine proteases and their inhibitors

Abstract: The data indicate that the cysteine cathepsins and their inhibitors are involved in a process related to early meningioma recurrence, regardless of their histological classification. Of note, the known tumor invasiveness marker cathepsin B, measured in whole-tumor homogenates, was not prognostic, in contrast to its endogenous inhibitor stefin B, which was highly significant and the only independent prognostic factor to predict meningioma relapse in multivariate analysis and reported herein for the first time. … Show more

“…Glioma cells were positioned close to the long retinal axons obtained derived from explants of embryonic chickens. Migration of cancer cells has been monitored by confocal microscopy and high-resolution video microscopy [255]. Overall, the invasiveness results obtained by confrontation assay are correlated with malignancy in vivo [256].…”

“…The advantages consist in a better representation of the invasion process, due to the fluorescent tag applied to the tumor cells, as well as in a better representation of extracellular matrix molecules normally encountered by invading glioma cells [133,222]. Oellers and collaborators described a similar refined protocol [255], presenting a co-culture of glioma cells with myelinated axons in vitro, with the aim to study the interactions between migrating glioma cells and nerve fibers. Glioma cells were positioned close to the long retinal axons obtained derived from explants of embryonic chickens.…”

“…Further studies are required to clarify the significance of the appearance of Rosenthal fibers. No difference in MIB-1 staining index was seen between cases of true and pseudo-invasion, and the question of whether any sort of difference in cellular activity exists between the two remains a matter for further study, although there are several reports up to now [5] [6].…”

“…Desmosomes are crucial for tissue homoeostasis and cardiac integrity . The cysteine protease inhibitors stefins have been proposed as prognostic and diagnostic tools for cancer and may have an important role in desmosome‐mediated cell–cell adhesion . The role of this regulatory family in the heart is unknown and warrants further investigation.…”
