Abstract: We previously demonstrated that the TNF-α-induced inside-out signaling leading to β2 integrin activation is redox regulated. To identify kinases involved in this pathway, the effects of kinase inhibitors on the expression of β2 integrin activation neoepitope (clone 24) were investigated. We show that both p38 MAPK (inhibited by SB203580) and Src kinases (inhibited by PP2) are involved in β2 integrin activation by TNF and oxidants in human neutrophils. Src kinases appeared constitutively active in resting neutr… Show more

“…These results, together with our previous observation illustrating that HP-NAP up-regulated the expression of ␤ 2 integrins (27), definitively confirm that HP-NAP has a direct role in stimulating neutrophils to adhere. Finally, we demonstrated the involvement of the p38-MAPK along the signaling cascade responsible for the integrin activation, as already reported for TNF-␣ (38).…”

“…Bouaouina et al (18) demonstrated that constitutively active Src TKs in neutrophils stimulated with TNF-␣ are involved in the integrin signaling that leads to ␤ 2 integrin activation in neutrophils. In contrast, the findings of the present study suggest that the inducible active Src TKs are involved in the integrin signaling activation induced by LPS.…”

“…For example, c-Src has been shown to directly phosphorylate proteins downstream of integrin signaling (e.g., focal adhesion kinases (FAK), Cas, and paxillin) following integrin ligation (17). Moreover, recently, Src TKs were demonstrated to participate in both "inside-out" signaling leading to integrin activation and to "outside-in" signaling as a result of interactions between activated integrins and the ECM (18).…”

“…The precise mechanism of integrin overexpression and airway inflammation among ragpickers is currently unknown. It is possible that microorganisms or their products present in MSW are involved, because bacterial endotoxin 15) and viruses 21) mediate TNF-α release from macrophages 22) which, in turn, mediate release of IL-8 from airway epithelial cells that attract neutrophils in the airways 23) . Neutrophils respond to IL-8 via receptors of this chemokine leading to chemotaxis, shape change and CD11b upregulation.…”
