Leptospirosis is a widespread zoonosis characterized by multiple organ failure and variable host susceptibility toward pathogenic Leptospira strains. In this study, we put the role of inflammatory mediators in parallel with bacterial burdens and organ lesions by comparing a susceptible animal model, the hamster, and a resistant one, the Oncins France 1 (OF1) mouse, both infected with virulent Leptospira interrogans serovar Icterohaemorrhagiae strain Verdun. Histological observations evidenced edema, congestion, hemorrhage, and inflammatory infiltration in the organs of hamsters, in contrast to limited changes in mice. Using reverse transcription-quantitative PCR techniques, we showed that the relative Leptospira burden progressively increased in hamster tissues, while a rapid clearance was observed in mouse tissues. The early regulation of the proinflammatory mediators interleukin-1␤ (IL-1␤), IL-6, tumor necrosis factor alpha, and cyclo-oxygenase-2 and the chemokines gamma interferon-inducible protein 10 kDa/CXCL10 and macrophage inflammatory protein-1␣/CCL3 in mouse tissues contrasted with their delayed and massive overexpression in hamster tissues. Conversely, the induction of the anti-inflammatory cytokine IL-10 was faster in the resistant than in the susceptible animal model. The role of these cytokines in the pathophysiology of leptospirosis and the implications of their differential regulation in the development of this disease are discussed.

Aims and Objectives: In the past decade, Leptospirosis has emerged as a globally important emerging infectious disease. Leptospires have several chemical components such as Lipoproteins, glycolipoproteins which are toxic and participate in the pathogenesis of the disease. This study was focused to find out the immune response of outer membrane proteins (OMPs) of Leptospira interrogans serovar tarassovi by estimating the cytokine profile and antibody levels from cases of Leptospirosis.Materials and Methods: A total of 120 blood samples were collected from patients (90 male/30 female) presenting with acute febrile illness and fulfilling the criteria of clinical diagnosis of Leptospirosis. All the 120 samples & 40 healthy controls were subjected to ELISA (IgM serion), MAT and Leptospira interrogans serovar tarassovi IgM ELISA and also to detect TNF? and IL-6 levels using (BD Opt EIA Human IL-6 & TNF? ELISA kits).Results: Sera which were found to be reactive in L. interrogans serovar tarassovi OMP IgM ELISA had significant levels of TNF? and IL-6 and antibody titres in ELISA and MAT.Conclusions: Patients from Group I had significant levels of TNF? in the serum i.e., p = 0.02 which indicates the pro-inflammatory Th1 response as TNF? are macrophage derived cytokine mediators. The IL-6 level of Group I, II, III were significantly higher than the Group IV (p = 0.02) suggesting that Th2 anti-inflammatory response seen in majority of these cases which has a protective role in leptospirosis.Asian Journal of Medical Sciences Vol.7(1) 2015 93-95

Background: Oral candidiasis is the most prominent oral fungal infection with Candida albicans (C. albicans) as 75% of ethiologic factor. Golden sea cucumbers (Stichopus hermanii) have been consumed by Asian community as folk medicine. It has been known to have antifungal and immunomodulator agent thus potential to be explored as treatment in oral candidiasis. Purpose: The aim of this study was to examine the potency of Stichopus hermanii extract as oral candidiasis treatment. Method: The study was an experimental laboratories research with post test only control group design. Thirty male wistar rats were divided into 5 groups i.e negative control, positive control and 3 treatment groups. Oral candidiasis condition were induced by spraying C. albicans suspension on dorsal tongue of wistar rats, once in 2 days for 14 days. The treatment groups were given Stichopus hermanii extract on the dose of 4.25 ml/kgBW, 8.5 ml/kgBW, 17 ml/kgBW once daily for 14 days. The expression of anti C. albicans antibody and TNF-α were examined by immunohistochemistry on ephitelial tongue. Data was analyzed by Manova and LSD test. Result: Anti C. albicans antibody expression were higher in positive control group than in negative control group while TNF-α expression were lower in positive control group than in negative control group (p<0.05). Treatment with Stichopus hermanii extract on all doses decreased the expression of anti C. albicans antibody and increased the expression of TNF-α (p<0.05). Conclusion: Stichopus hermanii extract decreased the expression of anti C. albicans antibody and increased the expression of TNF-α in epithelial rat tongue.
