Three-dimensional growth as multicellular spheroid activates the proangiogenic phenotype of colorectal carcinoma cells via LFA-1-dependent VEGF: implications on hepatic micrometastasis

Abstract: Background: The recruitment of vascular stromal and endothelial cells is an early event occurring during cancer cell growth at premetastatic niches, but how the microenvironment created by the initial three-dimensional (3D) growth of cancer cells affects their angiogenesis-stimulating potential is unclear.

“…Pretreatment of C26 cells with 2 lg/mL anti-murine LFA-1 antibodies for 45 minutes prior to coincubation with LSECs resulted in the same effect as ICAM-1 blockade. Anti-murine LFA-1 antibodies also decreased cancer cell attachment to LSECs, 19 suggesting that those C26 cells interacting with LSECs through other adhesion mechanisms were not involved in ManR and IL-1 up-regulation.
