Thermotoga neapolitana bgIB gene, upstream of lamA, encodes a highly thermostable β-glucosidase that is a laminaribiase

Abstract: The gene for thermostable 1,3-/?-glucosidase BglB was cloned from the chromosome of Thermotoga neapolitana and its primary sequence was determined. The purified recombinant pglucosidase B had a monomer molecular mass of 81 kDa in accordance with the amino acid sequence predicted from the nucleotide sequence of clone pTT51. It was a member of glycosylhydrolase family 3 and belonged to enzyme class EC 3.2.1.21. pGlucosidase B had a specific activity of 255 U mg-l on Lbnitrophenyl(PNP)-pglucoside a t the optima o… Show more

“…A schematic conversion of cellooligomers to activated glucosyl molecules. Enzymes indicated are CelA and CelB, endoglucanases (7) active in or on the toga; GghA, glucan glucohydrolase, and CbpA, cellobiose phosphorylase, intracellular enzymes (this work); and LamA and BglB, laminarinase and ␤-glucosidase, respectively (45,46). of the process of acquisition of carbon and energy by Thermotogales.…”

“…6). Other potential components of this catabolic pathway might include a laminarinase (LamA) and a laminaribiase/␤-glucosidase (BglB), which act on both ␤-1,3 and ␤-1,4 linkages (45,46).…”

“…The b-1,3-glucanases produce a characteristic pattern of b-1,3-oligosaccharides, which are further degraded by b-glucosidases. In contrast to the highly specific b-glucanases, b-glucosidases degrade laminarioligosaccharides (b-1,3) as well as cello-oligosaccharides (b-1,4) (Gräbnitz, et al 1989;Zverlov et al, 1997b).…”

“…LamA (endo-␤-(1,3)-glucanase) was a recombinant protein from Thermotoga neapolitana expressed in Escherichia coli (a kind gift from Dr. Vladimir V. Zverlov, Institute of Molecular Genetics, Russian Academy of Science, Moscow, Russia) (27). Endo-␤-(1,6)-glucanase from Trichoderma harzianum was expressed in Pichia pastoris (a kind gift from Prof. Stanley Brul, Swammerdam Institute for Life Sciences, Amsterdam, The Netherlands) (28).…”
