The Use of Monoclonal Antibodies in Enzyme Linked Immunosorbent Assays for Detection of Antibodies to Bovine Viral Diarrhoea Virus

Abstract: Summary Two ELISA techniques were developed for the detection of antibodies to bovine viral diarrhoea virus (BVDV), an indirect and a competitive ELISA. The coating antigen was prepared from virus harvests which were 10 to 100 times larger than usual. This was achieved by culturing the cells in a growth medium with foetal calf serum devoid of IgG. The harvested BVDV was solubilized with detergent to increase the accessibility of antigens. By these modifications, the amount of antigen produced from one 120 cm2 … Show more

“…The animals were housed in a barn with a tied stall system at the Department of Obstetrics and Gynaecology, Swedish University of Agricultural Sciences, Uppsala, Sweden, and fed according to Swedish standards [42]. They were serologically tested and free from Neospora caninum [9] and bovine viral diarrhoea virus (BVDV) infections [18]. The care of the animals and the experimental design of this study were approved by the Local Animal Ethics Committee in Uppsala, Sweden.…”

“…Present serological techniques do not discriminate between antibodies produced in pigs after infection with HCV and BVDV. Competition ELISA procedures using MAbs as described by Juntti et al (1987) can be implemented to approach this problem. Synthetic peptides reactive exclusively with antibodies to specific pestiviruses offer another diagnostic possibility.…”
