The role of androgen receptor gene mutations in male breast carcinoma.

Abstract: Certain germline mutations (607Arg-Gln, 608Arg-Lys) in the androgen receptor gene have been associated with the occurrence of breast cancer in males suffering from partial androgen insensitivity. To assess whether somatic mutations in this gene could be detected in breast carcinoma, archival tumor tissue of males without clinical evidence of androgen insensitivity was screened for point mutations in the androgen receptor gene. DNA was retrieved by chloroform-phenol extraction from formalin-fixed, paraffin-embe… Show more

“…AIS is a disorder affecting sexual differentiation in variable severity, ranging from phenotypic females (complete AIS, CAIS) to defective spermatogenesis in otherwise normal males (partial AIS, PAIS, or minimal AIS, MAIS) depending on the type and localization of these mutations 24,25 . Haploinsufficiency of the AR gene due to complete or partial gene deletions or nonsense and frameshift mutations usually lead to complete AIS.…”

“…In a study of 13 male breast cancer patients, one was observed to carry a similar mutation [48]. In another small study of 11 male breast cancer patients [49], this mutation was not observed. These results suggested that the mutation may play a role in the development of breast cancer in some males.…”

“…In retrospect, the clinical features of our patients were consistent with AIS, including the distant history of ‘removal of the uterus and ovary’ in infancy in one case, and ‘congenital absence of the uterus’ in the other. The history of breast carcinoma in the latter patient is also noteworthy, because there appears to be an increased incidence of breast neoplasia in patients with AR mutations 8–10 . Other than the neoplastic risks, the diagnosis of AIS presents obvious clinical and psychological issues to the patient, and potentially to her siblings and other family members.…”
