The Role of Alpha 6 Integrin in Prostate Cancer Migration and Bone Pain in a Novel Xenograft Model

Abstract: Of the estimated 565,650 people in the U.S. who will die of cancer in 2008, almost all will have metastasis. Breast, prostate, kidney, thyroid and lung cancers metastasize to the bone. Tumor cells reside within the bone using integrin type cell adhesion receptors and elicit incapacitating bone pain and fractures. In particular, metastatic human prostate tumors express and cleave the integrin A6, a receptor for extracellular matrix components of the bone, i.e., laminin 332 and laminin 511. More than 50% of all … Show more

“…The gene set enriched in Arrb2-dependent Apc Δ14/+ mouse tumors includes numerous genes that promote the evolution toward malignant and invasive tumors. Indeed, in vitro and in vivo studies have linked the expression of Itga6, Foxc1, and Mmp7, -10, and -12 to increased tumor cell invasion, migration, and metastasis (25)(26)(27). Target genes of the Wnt signaling pathway such as Prox1, Sox4, Sox9, Lgr5, and Cldn2 are also involved in tumorigenesis and cancer progression (20,(28)(29)(30)(31).…”

“…The monoclonal GoH3 antibody to alpha6 detected a band at 87 kDa corresponding to the heavy chain predicted at 88 kDa (UniProt, Q61739) and a smaller band that migrates at 60 kDa. Cleavage has been previously reported for this molecule with the product migrating at ϳ70 kDa under nonreducing conditions (King et al, 2008). The polyclonal antibody against alpha7 detected a unique band at 129 kDa corresponding to the full-sized molecule (UniProt, Q61738; Echtermeyer et al, 1996); the monoclonal 5C6 antibody against alphaM detected a single band at 127 kDa corresponding to the full-size molecule (UniProt, P05555).…”

“…Cilengitide, a cyclic Arg-Gly-Asp peptide that inhibits αvβ 3 and αvβ 5 , is a promising antagonist of αv integrins. A recent randomized phase II trial assessed the efficacy of different doses of Cilengitide in metastatic hormone refractory PCa patients (20).…”
