The regulation of AMP-activated protein kinase by phosphorylation

Abstract: The AMP-activated protein kinase (AMPK) cascade is activated by an increase in the AMP\ATP ratio within the cell. AMPK is regulated allosterically by AMP and by reversible phosphorylation. Threonine-172 within the catalytic subunit (α) of AMPK (Thr"(#) was identified as the major site phosphorylated by the AMP-activated protein kinase kinase (AMPKK) in itro. We have used site-directed mutagenesis to study the role of phosphorylation of Thr"(# on AMPK activity. Mutation of Thr"(# to an aspartic acid residue (T1… Show more

“…AMPK activation can acutely regulate metabolism as well as chronically alter gene expression to the same effect. High cellular concentrations of ATP antagonize the effects of AMP on AMPK so that AMPK responds sensitively to the AMP-to-ATP ratio [4,6,8,17,19,20,22,23,27,28,34,41]. In the present study, we establish the presence of AMPK in the mitochondrion-rich, particularly ATP-dependent, parietal cell.…”

“…In this in vivo model, ATP appeared to play a minor role in antagonism of the AMP-activation effect. Activation of AMPK by AMP in vitro is markedly inhibited by 4 mM ATP [170]. The NMR data suggest that the total ATP in heart is not accessible to AMPK, or possibly in the cellular environment, the enzyme is less sensitive to the ATP inhibitory effect observed in vitro.…”
