The redox state of the ascorbate-dehydroascorbate pair as a specific sensor of cell division in tobacco BY-2 cells

Abstract: The effects of ascorbate (ASC) and dehydroascorbate (DHA) on cell proliferation were examined in the tobacco Bright Yellow 2 (TBY-2) cell line to test the hypothesis that the ASC-DHA pair is a specific regulator of cell division. The hypothesis was tested by measuring the levels of ASC and DHA or another general redox pair, glutathione (GSH) and glutathione disulfide (GSSG), during the exponential-growth phase of TBY-2 cells. A peak in ASC, but not GSH, levels coincided with a peak in the mitotic index. Moreov… Show more

“…Similarly to our study, one of the antioxidative enzyme-L-ascorbate peroxidase 1 (spot no 9) was accumulated after ME-inducing treatment in maize and rapeseed (Joosen et al 2007;Cordewener et al 2009;Uváčková et al 2012). Its role can be more complex as in various plant cell culture systems high ascorbate levels stimulate cell division (de Pinto et al 1999).…”

“…The material was ground in liquid nitrogen and homogenized with 1 mL of cold 5% meta-phosphoric acid at 4°C in a porcelain mortar. The homogenate was centrifuged at 20,000 g for 15 min at 4°C, and the supernatant was collected for analysis of glutathione and ascorbate (or its analogues) according to de Pinto et al (1999).…”
