The Primary Motor Cortex Stimulation Attenuates Cold Allodynia in a Chronic Peripheral Neuropathic Pain Condition in Rattus norvegicus

Abstract: Background: The primary motor cortex (M 1) stimulation (MCS) is a useful tool for attenuation of the peripheral neuropathic pain in patients with pharmacologically refractory pain. Furthermore, that neurological procedure may also cause antinociception in rodents with neuropathic pain. Cold allodynia is a frequent clinical finding in patients with neuropathic pain, then, we evaluated if an adapted model of neuropathy induced by chronic constriction injury (CCI) of the ischiadicus nervus (sciatic nerve) produce… Show more

“…An adapted model of chronic constriction injury (CCI) of the nervus ischiadicus was used to induce NP. A single ligature consisting of a chromic catgut suture (Bioline Fios Cirúrgicos Ltda., Brazil) was loosely placed around the right nervus ischiadicus (9)(10)(11)(12). Each animal, in turn, was anesthetized with a mixture of ketamine (União Química Farmacêutica Nacional, Brazil) and xylazine (Hertape/ Calier, Brazil) (92 mg/kg and 9.2 mg/kg, respectively, ip).…”

“…We have recently reported an improvement in this procedure in a laboratory animal model of NP, inducing CCI of the right nervus ischiadicus through a single loose ligature in an adaptation of the NP model described by Bennett and Xie in 1988 (9-11). The modified model produced sensory, affective, and cognitive disorders in rodents (9)(10)(11)(12). In addition, mechanical allodynia has been recorded twenty-one days after nervus ischiadicus CCI surgery (10,11).…”

“…There are functional and metabolic similarities between neuropathic pain and post-traumatic stress disorder, with the mPFC playing a key role in integrating the disorder and comorbidities (Feldman, 2004 ). There are reports in the literature showing in both human patients and laboratory animals that the presence of chronic pain causes changes in functional reorganisations in cortical and subcortical structures, including the mPFC (Apkarian et al, 2009 ; Baliki et al, 2006 ; Medeiros et al, 2019a , b , c ; Malvestio et al, 2021 ; Metz et al, 2009 ), dorsal thalamus (Apkarian et al, 2004 ), amygdaloid complex (Han et al, 2005 ; Ji et al, 2010 ), and anterior cingulate cortex (ACC) (Li et al, 2010 ).…”
