The OST‐complex as target for RNAi‐based pest control in Nilaparvata lugens

Abstract: RNAi-based pest control strategies are emerging as environment friendly and species-specific alternatives for the use of conventional pesticides. Because N-glycosylation is important for many biological processes, such as growth and development, the early steps of protein N-glycosylation are promising targets for an RNAi-based pest control strategy. Through injection of dsRNAs, the expression of the catalytic subunits of the oligosaccharyl transferase complex was efficiently silenced in nymphs of the notorious… Show more

“…trimming caused a peak of mortality at the transition from nymph to adult. These results confirm the crucial role of N-glycan attachment for insect growth and survival as was previously observed in the holometabolous insects T. castaneum and D. melanogaster (Spradling et al, 1999;De Schutter et al, 2019). However, some differences between the phenotypes in holometabolous and hemimetabolous insects were observed.…”

“…Similarly, RNAi-mediated silencing of the genes encoding for the enzymes involved in these essential steps causes high mortality in N. lugens. Blockage of the N-glycan attachment, by cosilencing of both Stt3 isoforms, resulted in high nymphal mortality (De Schutter et al, 2019). While RNAi of the glucosidases and interference with the high-mannose N-glycan…”

“…Another possible explanation is that due to the increased pressure by silencing three genes a compensation effect takes place. A similar observation was made when silencing the catalytic subunit of the N-glycosylation oligosaccharyl transferase complex, after silencing of one isoform, the expression of the other isoform increased (De Schutter et al, 2019). Nevertheless, it is interesting that all double or triple RNAi experiments involving TcOGT resulted in a significantly higher larval mortality than the single RNAi, while the larval mortality was only minor in the single RNAi of TcOGT.…”

“…In the majority of the surveyed publications, only one target gene was used per bioassay, but multiple studies used two (for examples see the literature 41,51,52 ) and up to four target genes in a single RNAi assay. 53 The top five target genes by number observed, Vha, Ace, chitinase (Cht), COPI coatomer (COP), and cellulase (GHF) represent 24.5% of the data.…”
