From a cloned library of Drosophila DNA, a recombinant phage was isolated which contained a nucleotide sequence homologous to both the histone gene and a movable genetic element, 297 [Saigo, K., Millstein, L. & Thomas, C.A., Jr. (1981) Cold Spring Harbor Symp. Quant. Biol. 45, 815-827]. We have determined the nucleotide sequences of the termini of 297 as well as its insertion site in the histone gene in the clone. Our results strongly suggest that (i) although 297 ends with typical long terminal repeats, integrated 297 is not bounded by 5' T-G...C-A 3', dinucleotides commonly found at the very ends of integrated forms of other similar genetic elements, and (ii) the insertion of 297 occurred exactly within the T-A-T-A box for the H3 histone gene containing T-A-T-A-T-A. The structures of the insertion sites of 297 were also examined by using two other recombinant clones of the histone gene. A 6-base-pair-long segment, T-A-T-A-T-A, was again found to be present at the insertion sites of 297 in both clones. Based on these results, it was concluded that, at least within the histone gene cluster, the insertion of 297 was carried out in a site-specific fashion with T-A-T-A-T-A as the target sequence.

The organization of the 5S RNA gene cluster of Drosophila melanogaster is different in two Oregon R stocks that have been separated for a number of years. The Oregon R Yale population contains various different arrangements of the cluster. One of these is due to the insertion of a B104 element near one end of the cluster. Other arrangements lack the B104 insertion and have instead a variety of deletions originating in the vicinity of the B104 insertion site and removing from 0 to 60% of the 5S RNA genes without affecting nearby tRNA genes. In contrast, the Oregon R Heidelberg population has no B104 element in the 5S gene cluster and no heterogeneity in the arrangement of the cluster. We propose that transposable elements inserted at a genomic locus generate heterogeneity in a population at that locus due to excision of the element with and without accompanying deletions of flanking sequences. As a consequence, a fly population would accumulate a large number of deletions scattered throughout the genome in as many loci as contain transposable elements. We show further that D. melanogaster contains a large redundancy of 5S RNA genes since the 60% deletion of the cluster shows no visible phenotype when homozygous or when heterozygous against a total deletion of the entire 5S gene cluster.

Histone genes of the fission yeast Schizosaccharomyces pombe were cloned from Charon 4A and cosmid gene libraries by hybridization, and their nucleotide sequences were determined. The genome of S. pombe has a single, isolated H2A, a pair of H2A-H2B and three pairs of H3-H4 (one H2B, two H2A and three each of H3 and H4). This non-assorted histone gene organization is distinct from that of the budding yeast which has two pairs of H2A-H2B and H3-H4. The predicted amino acid sequences of S. pombe histone H2As, H3s and H4s were identical except for three residue changes in H2As. Compared with those of S. cerevisiae and human, variable residues were clustered near the NH2-and COOH-terminal regions of H2A and H2B. Sequence homologies to the two organisms were roughly the same in H2A (79-83%), H3 (92-93%) and H4 (91%), but differed in H2B (82% to S. cerevisiae and 68% to human). The coding sequences in the pairs of S. pombe histone genes were divergently directed. A 17-bp long highly homologous sequence (AACCCT box) that had internal 6-bp direct repeats was present in the intergene spacer sequences or in the 5' upstream region of all the cloned histone genes. A possible regulatory role of the common upstream sequence for histone gene expression is discussed.
