The Molecular Properties of the Methaemoglobin of Chironomus plumosus L. in Solution

Abstract: I. By means of an analytical ultracentrifuge the molecular weight of purified methaemoglobin from Chironomus plumosus has been determined a t different pH values. Under weak acidic conditions (pH 5.2 to 7.0) the sedimentation constant amounts to 2.73 & 0.02 S and the diffusion constant varies from 7.5 to 8.5 F. The molecular weight was calculated as 32,000 & 1,500. 2.Outside of this pH-range the molecular weight decreases. I n alkaline solution (pH 9.2-10.0) the dimer molecule splits off reversibly into monome… Show more

“…In C plumosus and C th ummi, both monomeric (mol wt '" 16,(00 ) and dimeric forms of hemoglobin occur (Table 2), dissociation being favored at elevated pH, although the situation in vivo is not certain (339,341,(378)(379)(380). In C tentans and some other species, however, only the monomers are fo und ( Table 2; 93).…”

“…Furthermore, pHdependent association-dissociation equilibria have been observed with Chirononaus plumosus haemoglobins not separated chromatographically [15,3]. The Bohr effect and the shape of the 0,-binding curves might be related to these equilibria of aggregation.…”
