The long noncoding RNA lncNB1 promotes tumorigenesis by interacting with ribosomal protein RPL35

Abstract: The majority of patients with neuroblastoma due to MYCN oncogene amplification and consequent N-Myc oncoprotein over-expression die of the disease. Here our analyses of RNA sequencing data identify the long noncoding RNA lncNB1 as one of the transcripts most over-expressed in MYCN-amplified, compared with MYCN-non-amplified, human neuroblastoma cells and also the most over-expressed in neuroblastoma compared with all other cancers. lncNB1 binds to the ribosomal protein RPL35 to enhance E2F1 protein synthesis, … Show more

“…In vitro enforced expression of RPL22L1 increased tumorigenicity and mesenchymal markers. In our analysis, RPL35 was also associated with MNA tumours and reportedly plays an important role in neuroblastoma [23]. Specifically, RPL35 protein bound lncNB1 (a long non-coding RNA) and E2F1 mRNA and facilitated E2F1 mRNA translation into protein, leading to increased DEPDC1B gene transcription.…”

“…The binding of IGF2BPs to RNA relies on the m 6 A RNA epi-transcriptomic modification [ 58 ]. All family members are reported to be expressed in up to 100% of neuroblastoma tumours, with high IGF2BP1 expression associated with poor overall prognosis and MYCN amplification [ 59 ], as well as MNA cell lines [ 23 ]. In our analyses, IGF2BP1 was highly expressed in MNA tumours, though the significance was lower than the top elucidated candidates discussed here.…”

“…LncRNAs suppress (Rinn et al, 2007;Kaneko et al, 2014;Kim et al, 2018) or activate (Wang et al, 2011;Gomez et al, 2013;Luo et al, 2019; gene transcription by binding to chromatin remodeling protein complexes and by acting as transcriptional enhancers (Lai et al, 2013;Jiang et al, 2018). In addition, lncRNAs post-transcriptionally regulate mRNAs, such as pre-mRNA alternative splicing (Beltran et al, 2008;Andersen et al, 2019), mRNA stability (Yang et al, 2014;Liu et al, 2014), and translation into proteins (Yoon et al, 2012;Malakar et al, 2019;Liu et al, 2019).…”

“…RPS20 was identi ed as a new oncogene in glioblastoma [30] and colon cancer [31]. RPL35 was identi ed as an key factors for tumorigenesis in neuroblastoma [32]. RPS2 was overexpressed in malignant prostate cancer and had the potential to be therapeutic target for prostate cancer [33].…”
