The Isolation and Selected Properties of Blood Monocytes

Abstract: A technique is described for the quantitative recovery of monocytes from horse blood by means of flotation on dense albumin solutions. Monocytes are concentrated in a surface pellicle along with a few lymphocytes which are then removed when the monocytes adhere to a glass surface. The in vitro cultivation of homogeneous populations of monocytes results in an increase in (a) cell size, (b) number of mitochondria, and (c) phase-dense granules of the centrosphere. The phase-dense granules are osmiophilic and acid… Show more

“…Such observations have been confirmed repeatedly (4,5) and extended to document increases in cell size, protein content, lysosomal and ectoenzyme activity, surface receptor expression, and phagocytic capacity (4,(6)(7)(8)(9)(10)(11)(12). This differentiation sequence in vitro closely parallels that observed when monocytes emigrate from the vasculature into the tissues (13)(14)(15).…”

“…The albumin gradient, a modification of the method of Bennett and Cohn (22), yielded 15% more monocytes than did Ficoll-Hypaque but no significant differences were noted in either cell morphology or in vitro cell survival. This monocyte yield (74%) equalled the maximum previously reported for other albumin gradients (30-75%) (10,11) and exceeds the estimated 50% yield by the method of Einstein et al (6).…”

“…The process of the in vitro transformation of blood monocytes to mature macrophages has been extensively studied by different investigators using mainly monocytes from animal sources (2,16,33,35), and increases in lysosomal enzyme activities have been reported. Similar in vitro changes have also been described in cultured mouse peritoneal macrophages (16).…”
