The Hoorn Diabetes Care System (DCS) cohort. A prospective cohort of persons with type 2 diabetes treated in primary care in the Netherlands

Abstract: PurposePeople with type 2 diabetes (T2D) have a doubled morbidity and mortality risk compared with persons with normal glucose tolerance. Despite treatment, clinical targets for cardiovascular risk factors are not achieved. The Hoorn Diabetes Care System cohort (DCS) is a prospective cohort representing a comprehensive dataset on the natural course of T2D, with repeated clinical measures and outcomes. In this paper, we describe the design of the DCS cohort.ParticipantsThe DCS consists of persons with T2D in pr… Show more

“…Individuals who participated in this study are part of the Hoorn DCS cohort, a prospective cohort of over 12,000 individuals with type 2 diabetes [ 16 ]. People visit the DCS annually for routine care and data collection, including anthropometric, fasting glucose, HbA 1c , blood lipid and blood pressure measurement and information on medication use.…”

“…Those that have been conducted have tended to be small cross-sectional case–control studies. We have investigated the relationship between blood gene expression levels and HbA 1c levels in almost 400 individuals with type 2 diabetes selected from the Hoorn Diabetes Care System (DCS) cohort [ 16 ].…”

“…In total, 3089 persons with T2D from 5 independent Dutch cohort studies, the Hoorn Diabetes Care System (DCS) West-Friesland (19), the Maastricht study (20), the Rotterdam study (RS) (21), the Netherlands Epidemiology of Obesity (NEO) study (22), and the Cohort of Diabetes and Atherosclerosis Maastricht study (CODAM) (23) were included. The selection processes of the independent studies have previously been described in detail; a brief description of the selection from each cohort for the present study, is provided in the following paragraphs.…”

“…The DCS provides diabetes care to people with T2D living in the West-Friesland region, who yearly visit the DCS research center (19). At the yearly visits, a medical exam is performed, and blood is drawn for biochemistry.…”

“…The final set of 2622 SNPs was constructed by overlapping the ExAC based and RNA-Seq based SNP lists. Both 2622 SNP and 50 SNP panels were used to evaluate the ability for sample and mismatch identification in samples from the Diabetes Care System (DCS) cohort [ 40 ], where there were 562 RNA-Seq as well as 3428 GWAS samples imputed using the HapMap reference panel. Genotype calling at all genomics coordinates was performed for the two SNP panels in both RNA-Seq and imputed GWAS samples.…”
