Abstract: Store-operated calcium channels provide calcium signals to the cytoplasm of a wide variety of cell types. The basic components of this signaling mechanism include a mechanism for discharging Ca2+ stores (commonly but not exclusively phospholipase C and inositol 1,4,5-trisphosphate), a sensor in the endoplasmic reticulum that also serves as an activator of the plasma membrane channel (STIM1 and STIM2), and the store-operated channel (Orai1, 2 or 3). The advent of mice genetically altered to reduce store-operate… Show more

“…Great progress has since been made and it is now well established that ER luminal Ca 2+ decrease causes STIM1 molecules to change their conformation such that they undergo massive clustering in the ER and interact and activate the Orai1 channels in the PM. Several important reviews have summarized these advances (Hogan et al 2010;Soboloff et al 2012;Hogan & Rao, 2015;Derler et al 2016a;Putney et al 2017;Zhou et al 2017). More recent studies have revealed structural features of the STIM1 and Orai1 proteins that are critical for the activation process.…”

“…STIM-ORAI ( Stim - Orai is used here for gene or gene products in rodents) interactions enable sustained Ca 2+ influx in many cell types and represent the SOCE prototype [42, 43]. A study analyzing the expression pattern of ORAI proteins in mouse dental tissues found ORAI1 expression in pre-ameloblasts during the late embryonic (E17) and early postnatal stages, with relatively high expression during the secretory stage [44].…”

“…Store-operated Ca 2+ entry regulates a diverse set of signaling pathways and is essential in virtually all eukaryotes (Prakriya and Lewis 2015; Amcheslavsky et al 2015; Putney et al 2016). Transient depletion of ER Ca 2+ stores is sensed by the Ca 2+ -binding EF hands of the single-pass transmembrane protein STIM1 (Soboloff et al 2012).…”
