The effect of storage on the expression of platelet membrane phosphatidylserine and the subsequent impacton the coagulant function of stored platelets

Abstract: These results show that PS expression increases after preparation of PCs from platelet-rich plasma and rises progressively during platelet storage under blood bank conditions. Furthermore, the greater PS expression is associated with increased platelet- dependent thrombin-generating capacity.

“…Possible effectors of the immunosuppressive side effects of PLT transfusions are the large amounts of PLT ectosomes (PLT-Ecto) that are released during storage and transfused concomitantly with PLT (18,19). Placke et al (20) showed that PLT shed and transfer their MHC class I molecules to tumor cells, conferring them resistance to NK cells.…”

“…a potential haemostatic advantage of stored red cell and platelet units may be explained by the observation that during storage red cells and platelets showed a progressive increase in membrane phosphtidylserine expression that correlates with increased thrombin generation [17]. Furthermore, platelet storage is associated with microparticle formation that has a potent effect on thrombus generation and propagation [18].…”

“…But this platelet staining method needs to perform an incubation of the dye with platelet in vitro. The routinely used procedures to prepare platelet-rich plasma are based on centrifugation (16)(17)(18). It has been recently demonstrated that such procedures lead to changes of platelets characteristics and specially concerning the expression of surface markers.…”
