Abstract: Quinazolines are multitarget agents, which have broad spectrum of biological activity, and some of them are now in cancer clinical testing. 3-(5-nitro-2-thienyl)-9-chloro-5-morpholin-4-yl[1,2,4]triazolo[4,3-c]quinazoline is a new synthetically prepared derivative, which in our previous study showed cytotoxic effects on cancer cell lines HeLa and B16. Quinazoline, at micromolar concentrations, induced morphological changes and necrosis of B16 cells, and at nanomolar concentrations it produced changes of F-actin… Show more

“…22 Activation of p38 MAPK and JNK is also related to increased activity of classical apoptotic mediators such as p53 and caspase 3 in different cells, such as cardiac myocytes and murine leukemia L1210 cells. 23,24 In non-stressed cells, p53 is in a latent form and is constitutively repressed by the binding of two proteins, Mdm2 and the inactive form of JNK, which mediate the degradation of p53 by the proteasome. 25 In response to stress, p53 phosphorylation coincides with its dissociation from both Mdm2 and JNK.…”

“…A number of photoprotective compounds such as scytonemins, mycosporines, mycosporine-like amino acids, phenylpropanoids and flavonoids (higher plants), melanins (humans and other animals and even some bacteria), and several other UV-absorbing substances of unknown chemical structures from different organisms have been developed to counter the photodamage. , 1,2,3-Triazoles have gained special attention because of their potent applications ranging from medicinal, materials, and biological research areas. More importantly, fused triazoles are of interest because of their various biological activities and clinical applications. , 1,2,3-Triazoles such as biscotrizole and drometrizole, which absorb UVA and UVB radiation, are used as active ingredients for the application of skin photoprotection. On the basis of this background, our research group has synthesized a novel series of bicyclic[1,2,3]triazoles as photoprotective agents that absorb light in the UVA region, and we have delineated their effect on UVA-induced senescence in human dermal fibroblast cells (FB) and the associated mechanism.…”

“…3). In our preliminary experiments [23], NTCHMTQ induced concentration-and time-dependent increase in caspase-3 activity. As presented in Fig.…”

“…In addition, reduction of tumor volume (~11.2 %) in intramuscular transplanted B16 cells was observed on day 16 at doses of 50 mg/kg. Interestingly, NTCHMTQ induced the increase of the sub-G 0 cell fraction, induced apoptotic DNA fragmentation and morphological changes and activated caspase 3 in murine L1210 leukemia cells [21][22][23]. These results demonstrated that NTCHMTQ is potent inducer of apoptosis in L1210 cells and futher studies on the molecular mechanism regulating the NTCHMTQ-induced apoptosis are necessary.…”
