The effect of 2,4-dichlorophenoxyacetic acid (2,4-D) on callus growth and production flavonoid content on culture callus Acalypha indica L.

Abstract: The Objectives of the research were to study the effect of 2,4-dichlorophenoxyacetic acid (2,4-D) on callus initiation and growth, as well as on flavonoid content on Acalypha indica callus. The outline of the research was that Acalypha indica as a medicinal plant potentially to be developed by in vitro culture method. The addition of 2,4-D in culture medium could induce cell proliferation, meanwhile, it also affects callus growth and production. Completely randomized design with three replicates was used in th… Show more

“…The use of different ZPT gives a variety of responses to each explant individual. This is due to the interaction between endogenous hormones in plant tissue and given exogenous growth regulators [13].Addition of 2.4 D in the media will stimulate cell division and enlargement in the explants so as to stimulate callus formation [14]. The addition of BAP into the media was able to play an active role in the growth and ploriferation of callus [15].…”

“…Growth regulating agent kinetin works in the regulation of protein synthesis which can accelerate cell division to result in callus growth. The substance that regulates the growth of auxin or 2,4-D [26], works to spread the cell wall so that the hydrogen bond is released. The release of hydrogen bonds will cause the entry of water into the cell, the pressure of the turgor will rise which will cause cell enlargement…”

“…Perbedaaan kuantitas kalus terjadi karena eksplan tangkai daun yang digunakan memiliki kepekaan dan daya serap terhadap media yang berbeda serta adanya pengaruh dari zat pengatur tumbuh yang diberikan. Kecepatan sel dalam membelah akan mempengaruhi volume sel (Rahayu et al, 2003). Perlakuan terakhir yang belum menunjukkan adanya tanda-tanda berkalus adalah perlakuan B3N1.…”
