The Cystic Fibrosis Upper and Lower Airway Metagenome

Abstract: Cystic fibrosis (CF) is the most common life-limiting monogenetic disease in European populations and is caused by mutations in the CFTR gene. Chronic airway infections with opportunistic pathogens are the major morbidity that determines prognosis and quality of life in most people with CF.

“…Moreover, the healthy control group was on average younger than the people with BE. However, as the gradient of depletion noted in patients with BE matches with that observed in patients with cystic fibrosis seen at the Hannover clinic [ 12 ], the divergent geographic origin of samples from Scotland and Northern Germany should not have substantially influenced the outcome of our study.…”

“…The strongest depletion was seen for the phylogenetically unrelated species F. periodonticum , E. sulci and N. subflava. We have observed the same pattern of depletion in sputa collected from individuals with cystic fibrosis [ 12 ]. F. periodonticum and E. sulci have so far not been investigated in the context of respiratory health and disease.…”

“…In comparison for the oral cavity, there was a significant positive correlation between plasma IL-8 and the genera Porphyromonas and Alysiella . Given that Staphylococcus aureus and Stenotrophomonas maltophilia are opportunistic pathogens in the respiratory tract and frequently detected in cystic fibrosis patients, it may be speculated that innate immune function in the lung mucosa could contribute to shifts in these genera [ 54 ]. The pathogenicity of Alysiella is unknown but it is commonly found in the oral cavity of mammals, whereas Porphyromonas gingivalis is associated with periodontal disease [ 55 , 56 ].…”
