The characteristics of bone marrow-derived endothelial progenitor cells and their effect on glioma

Abstract: BackgroundEPCs were isolated primarily in 1997 by Asahara et al. and recent studies indicated that bone-marrow-derived EPCs contributed little to the endothelium of tumor vessels. Tumors of the CNS system demonstrate various features of angiogenesis.MethodsEPCs derived from rat bone marrow were isolated and cultured in M199 medium without any induced factors. EPCs were studied using immunohistochemical staining, Flow cytometry and culture under three-dimensional condition to determine EPCs’ characteristics in … Show more

“…These cells were called late EPCs, and we used late EPCs for further studies. EPCs expressed surface markers, such as CD31 (96.8±0.5%), CD34 (97.2±1.4%), CD133 (92.8±5.3%) and VEGFR-2 (91.5±3.7%) (Figure 1C), and this finding is consistent with previous studies [20, 21]. EPCs could differentiate into adipocytes (Figure 1D) and smooth muscle cells (Figure 1E).…”

“…In brain tumors, soluble factors released by glioma-associated myeloid cells (GAMs; mainly macrophages and microglia), promote cell invasion and angiogenesis 18 , 19 . Different K + channels expressed by GAMs modulate phagocytosis, cell migration and proliferation 20 22 , with Kv1.3 regulating microglia cell proliferation and neurotoxicity 23 , 24 .…”

“…Briefly, EPCs were isolated from human umbilical cord blood and cultured in medium containing M199 supplemented with 200 ml/L FBS, 10 μg/L vascular endothelium growth factor (VEGF; eBioscience, USA), 15g/L bovine pituitary extract, and 10 μg/L basic fibroblast growth factor (bFGF; eBioscience, USA) as previously described [ 15 ]. To analyze the function of EPCs, adherent cells were first incubated with 2.4 mg/L of DiI-acetylated low-density lipoprotein (Dil-acLDL) (MolecularProbe, USA), then stained with FITC-Ulex europaeus lectin-1 (FITC-UEA-I) (Sigma, USA).…”
