The Bewildering Effect of AMPK Activators in Alzheimer’s Disease: Review of the Current Evidence

Abstract: Alzheimer’s disease is a multifactorial neurodegenerative disease characterized by progressive cognitive dysfunction. It is the most common form of dementia. The pathologic hallmarks of the disease include extracellular amyloid plaque, intracellular neurofibrillary tangles, and oxidative stress, to mention some of them. Despite remarkable progress in the understanding of the pathogenesis of the disease, drugs for cure or disease-modifying therapy remain somewhere in the distance. From recent time, the signalin… Show more

“…This effect is relevant for cognition, since long-term memory requires new protein synthesis (Alberini, 2008). The effects of AMPK on amyloid β peptide accumulation, tau aggregation, and oxidative stress have been recently reviewed (Assefa et al, 2020). The role of AMPK in HD and ALS is discussed later in the appropriate sections.…”

“…For AD, the effects of AMPK activators remain controversial. As reviewed recently by Assefa et al ( 2020 ), most AMPK activators evaluated in AD are indirect activators (such as resveratrol and metformin). In addition to AMPK, these compounds also have other targets important for neuronal functions and survival.…”

“…AMPK plays a significant role in regulating the cell metabolism and energy status; and recent studies indicate that it may play a key role in AD (Assefa et al, 2020). Phosphorylation of AMPK at Thr( 172) is important for activation of the kinase (Stein et al, 2000).…”
