Sildenafil citrate has low oral bioavailability, systemic adverse effects, and a relatively delayed action. These issues may be addressed through direct transdermal delivery to the penis. This study aims to investigate the microemulsion formulation of the drug for effective transdermal delivery. Sildenafil citrate was formulated as a microemulsion using clove oil, dimethyl sulphoxide, phosphate buffer (pH 7), propylene glycol, Tween ® 80, and distilled water. Different proportions of these components were used to create six formulations of the microemulsion (F1-F6), which were then characterised by their physical appearance and clarity, pH, viscosity, conductivity, percent transmission, and droplet size. Furthermore, the stability, content analysis, in-vitro drug release, and transdermal permeation of sildenafil citrate from the generated drug-loaded microemulsions were studied. All prepared formulas contained nano-sized oil droplets (less than 20 nm), and the pH values were within the range of skin pH; however, two formulas were not transparent. Additionally, all formulations were thermodynamically stable, passing freeze-thaw, heating-cooling, and centrifugation tests. Next, the formulas demonstrated zero-order release kinetics, indicating that they can provide a sustained release profile for sildenafil citrate. Finally, the microemulsion formulation exhibited a 2.8-fold enhancement in skin permeation compared with that of the sildenafil citrate suspension. The prepared microemulsions demonstrated beneficial physical properties and skin permeation profiles that are promising for the local administration of sildenafil citrate.

Rheumatoid arthritis (RA) is a debilitating disease associated with locomotion impairment, and conventional therapeutic drugs are not optimal for managing RA. There is an avalanche of medications used for the management of RA. Still, studies have shown that they are associated with severe side effects, including hepatotoxicity, retinopathy, and cardiotoxicity disorders of the central nervous system (CNS), skin, blood, and infections. Complementary and alternative medicine (CAM) is currently gaining attention as a novel panacea for managing debilitating diseases, such as RA. Nigerian folk herbal remedies are replete with a plethora of curative medicine, albeit unvalidated scientifically but with seemingly miraculous provenance. Studies of the identification of bioactive compounds present in these botanicals using advanced spectral analytical techniques have enhanced our understanding of the role of Nigerian herbal remedies in the treatment and management of RA. Interestingly, experimental studies abound that the bioactive compounds present in the extracts of plant botanicals protected animals from the development of RA in different experimental models and reduced the toxicity associated with conventional therapeutics. Validated mechanisms of RA amelioration in human and animal models include suppression of the expression of NF-κB, IL-1β, TNF-α, IL-6, IL-8, IL-17, IL-23, chemokines, TGF-β, RANKL, RANK, iNOS, arginase, COX-2, VEGFA, VEGFR, NFATC1, and TRAP in the synoviocytes. Decreased ROS, NO, MDA, carbonyl groups, and PGE2 in the synovial fluid increased the expression of PPARα/γ; antioxidant and anti-inflammatory molecules also improve RA etiology. In this mini-review, we discuss the global burden of RA, the novel role of plant-based botanicals as potential therapeutics against signaling pathways in RA. Also addressed is the possible repurposing/reprofiling of plant botanicals to increase their therapeutic index among RA patients that patronize traditional healers in Nigeria with a global projection.

Essential oils are being used to improve health due to their high nutritive value. In this study, the comparative effects of Sesame oil and Clove oil were assessed on growth performance, blood profile and immunity of male rabbits. A total of 108 male rabbits were divided into 3 treatment groups; Treatment group A (Sesame oil), Treatment group B (Clove oil), Treatment group C (Sesame oil + Clove oil). Each treatment group was further divided into 4 subgroups according to dosage design of oils. The control group was untreated, while treatment dose for groups A and B was divided into 3 different concentrations; 0.1, 0.2 and 0.3ml/kilogram Body Weight. Whereas, the treatment concentrations for group C were as; 0.1+0.1, 0.2+0.2 and 0.3+0.3ml/kilogram Body Weight of oils for 1-1.5kg of rabbits. The rabbits were managed in proper caging under controlled conditions of photoperiod (12 light: 12 dark cycles) 23±2ºC of temperature. The selected dose concentration of each oil was administered orally and once a day for six weeks. The results showed that both oils had significantly enhanced the growth performance and reduced the Feed Conversion Ratio. The hematological parameters were increased in all treatment groups, respectively. The Enzyme-linked Immunosorbent Assay for Rabbit Hemorrhagic Disease Virus also showed a significant increase in antibody titer level. The study showed that Sesame oil and Clove oil as dietary supplements have significantly (p<0.05) improved overall performance of male rabbits. The current study clearly indicated that both oils could be a better choices for health purposes.
