Green chicory (Cichorium intybus) leaf is consumed worldwide that has tremendous bio-functional activities. This study was designed to elucidate the bio-functional activities of the green chicory leaf extract (CLE). Trolox equivalent antioxidant activities were carried out against DPPH, ABTS radical scavenging and ferric reducing activities. CLE showed remarkable hepatoprotective activity against oxidative damages and reactive oxygen species reducing effect in RAW 264.7 and Hep G2 cells. The anti-inflammatory effects performed on LPS-induced RAW 264.7 cells and primary spleen cells. In RAW 264.7 cells, Chicory leaf extract significantly restored the iNOS, COX-2, IL-6, TNF-α and other pro-inflammatory cytokines in mRNA expression. Meanwhile, it restored NF-κB (P-65) and MAPK pathways activation in immunoblot assay dose-dependently and repressed the P-65 translocation in the nucleus. Moreover, it showed notable anti-proliferative activities and downregulated NF-κB (P-65) and MAPK activation in LPS-stimulated splenocytes. Our findings suggest that CLE could be used as a natural anti-inflammatory agent. ARTICLE HISTORY

Citri reticulatae Pericarpium (CRP) is widely consumed as a food and herbal medicine in various countries and has many biological activities, including anti-inflammatory effects, but the molecular mechanism is not completely known. This study aims to investigate whether CRP extract and its major flavonoids hesperidin (Hsd) and hesperetin (Hst) can reduce inflammation in macrophages and the underlying mechanism.The chemical composition of CRP was detected by ultra-performance liquid chromatography-mass spectrometry (UPLC-MS) analysis. The inflammatory cell model was induced by stimulating RAW 264.7 cells with lipopolysaccharide (LPS, 1 μg/ml).Levels of nitric oxide (NO) and cytokines were determined by Griess assay and enzymelinked immunosorbent assay kits, respectively. Protein expressions were examined by western blotting. CRP extract dose-dependently suppressed LPS-induced production of NO, interleukin-6 (IL-6), and tumor necrosis factor-α. Elevated expressions of inducible NO synthase (iNOS) and cyclooxygenase-2 (COX-2) and phosphorylation of c-Jun NH 2 -terminal kinase, extracellular signal-regulated kinase, p38, and p65 in LPSstimulated cells were prevented by CRP extract. Hsd and Hst are flavonoids present in CRP, as confirmed by UPLC-MS analysis. Both Hsd and Hst ameliorated LPS-triggered inflammation to reduce NO release, IL-6 generation, and protein expressions of iNOS and COX-2. Advances in understanding of health‐promoting benefits of medicine and food homology using analysis of gut microbiota and metabolomics

The health-promoting benefits of medicine and food homology (MFH) are known for thousands of years in China. However, active compounds and biological mechanisms are unclear, greatly limiting clinical practice of MFH. The advent of gut microbiota analysis and metabolomics emerge as key tools to discover functional compounds, therapeutic targets, and mechanisms of benefits of MFH. Such studies hold great promise to promote and optimize functional efficacy and development of MFH-based products, for example, foods for daily dietary supplements or for special medical purposes. In this review, we summarized pharmacological effects of 109 species of MFH approved by the Health and Fitness Commission in 2015. Recent studies applying genome sequencing of gut microbiota and metabolomics to explain the activity of MFH in prevention and management of health consequences were extensively reviewed. We discussed the potentiality in future to decipher functional activities of MFH by applying metabolomics-based polypharmacokinetic strategy and multiomics technologies. The needs for personalized MFH recommendations and comprehensive databases have also been highlighted. This review emphasizes current achievements and challenges of the analysis of gut microbiota and metabolomics as a new avenue to understand MFH.
