The Anti-Inflammatory and Immunomodulatory Activity of Thymulin Peptide is NF-κB-Dependent and Involves the Downregulation of IκB-α

Abstract: The immunomodulatory activity of thymulin and related analogues in vivo is not well characterized in the CNS. We have previously provided evidence for an anti-inflammatory potential of thymulin in downregulating proinflammatory cytokines in a NF-B-dependent mechanism in vitro. Furthermore, we have shown that intracerebroventricular (ICV) treatment with thymulin in the hippocampus (HC) reduced the nuclear localization and activation of NF-B, an effect mediated by the IB-α/pIB-α pathway in vivo. ICV stereota… Show more

“…Literature data testify to the regulatory effects of thymulin on the activity of macrophages, the differentiation and functioning of T-lymphocytes [18]. Animals with a model of neuroinflammation show the stimulating effect of this hormone on IL-10 synthesis in the brain against the suppression of the synthesis of TNF-α and IL-1β [29]. Macrophages and T-lymphocytes have been shown to be the source of proinflammatory cytokines and active forms of oxygen that damage nerve cells of the brain [3,11,30].…”

“…Efficacy of such thymulin influence was lesser in old organism [27]. Antiinflammatory effect of thymulin in the brain tissue (production of proinflammatory cytokines) plays role in the maintenance of neurogenesis [28].…”

“…According to Oksimets et al [30], cytokines IL-1β, IL-2, IL-6, IL-8, TNF-α, which are secreted by transplanted MMSCs, play an important role in regulation of osteogenesis in injured limbs. The involvement of transcription factor NF-κB in implementation of thymulin effect in tissues in vivo and in vitro has been shown by altering the synthesis of proinflammatory (TNF-α, IL-1β, IL-6) and anti-inflammatory cytokines (IL-10) [31].…”
