Abstract: The adenylate cyclase toxin (CyaA) of Bordetella pertussis is a major virulence factor required for the early phases of lung colonization. It can invade eukaryotic cells where, upon activation by endogenous calmodulin, it catalyzes the formation of unregulated cAMP levels. CyaA intoxication leads to evident toxic effects on macrophages and neutrophils. Here, we demonstrate that CyaA uses the αMβ2 integrin (CD11b/CD18) as a cell receptor. Indeed, the saturable binding of CyaA to the surface of various hematopoi… Show more

“…To determine whether the cytokine modulatory effects of A-CyaA and NA-CyaA are mediated by their binding to cells via CD11b/ CD18, J774 macrophages were treated with the CyaA proteins and CpG-ODN in the presence of the anti-CD11b Ab M1/70, which has previously been shown to block the binding of A-CyaA to host cells (3). Addition of the anti-CD11b Ab suppressed the ability of A-CyaA and NA-CyaA to inhibit CpG-ODN-induced production of TNF-␣ and also their ability to synergize with CpG-ODN to enhance IL-10 secretion (Fig.…”

“…Differences in the acylation pattern have been demonstrated to alter the hemolytic and pore-forming activity of CyaA, but not its capacity to deliver the enzymatic domain into the cell cytosol (13,14). The channel-forming activity of CyaA exhibits a cooperative concentration-dependent curve, indicative of protein oligomerization, while intracellular cAMP accumulation can be triggered by protein monomers (3,15). These differences have led researchers to suggest that cell lysis and intracellular cAMP accumulation occur through separate mechanisms.…”
