Abstract: The process of bone turnover displays variations over 24 h, with C-terminal cross-linked telopeptide of type 1 collagen (CTX) and osteocalcin exhibiting a nadir in the afternoon and a peak in the night. In contrast, N-terminal propeptide of type 1 procollagen (P1NP) did not display an apparent 24-hour rhythm. Other emerging novel biomarkers of bone, sclerostin and Dickkopf-related protein 1 (DKK1), are markers of osteocyte activity with limited data available regarding their 24-hour profiles. In this study, we… Show more

“…Finally, there may be potential limitations to using DKK1 as a biomarker because of its large inter- and intraindividual variation over time, as demonstrated in a recent study examining 24-hour levels of DKK1 in healthy patient samples [32]. We do not expect this to be a major issue in our study because all samples were collected under the same conditions (in the morning following an overnight fast).…”

“…Furthermore, it has been is hypothesized that TSH plays a role in maintenance and repair mechanisms, including bone turnover ( 100 ). These processes fluctuate over time, with a bone resorption marker, C-terminal cross-linked telopeptide of type 1 collagen (CTX-I), exhibiting a strong circadian rhythm ( 101 ).…”

“…CTX, C-terminal cross-linked telopeptide of type 1 collagen, has been shown to have a circadian rhythm similar to osteocalcin with a low point in the afternoon and high point at night. No distinct rhythms were observed for PINP, sclerostin or Dkk1 [71]. The circadian clock becomes less synchronous with age.…”
