Background: To investigate the effects of taurine on prostate cancer cell proliferation and apoptosis, and on the mammalian sterilization of a 20-like kinase-1 (MST1)/Hippo signaling pathway. Methods:The prostate cancer DU145 cell line was selected and the 3-(4,5-dimethylthiazol-2-yl)-2,5diphenyltetrazolium bromide (MTT) method was used to determine the rate of inhibition by taurine on the proliferation of the cells at 1, 10 −1 , 10 −2 , 10 −3 , 10 −4 , 10 −5 and 10 −6 mg/mL to obtain the taurine intervention concentrations. The cultured cells were divided into three groups: the blank group was cultured with conventional culture medium, the positive control group was cultured with 2 mg/mL cisplatin, and the taurine group was cultured with the determined taurine intervention concentrations of 0.003 mg/mL as low, 0.03 mg/mL as medium and 0.3 mg/mL as high concentration. After 72 h incubation, cell proliferation, apoptosis and cellular MST1/Hippo signaling pathway protein expression were observed.Results: In the comparison of cell proliferation rate, the taurine group was lower than the positive control group and the blank group (P<0.05), the cell proliferation rate of different concentrations in the taurine group decreased with the increase of concentration (P<0.05). The apoptosis rate was higher in the taurine group than in the positive control group and the blank group (P<0.05), the apoptosis rate increased with increasing concentration in the taurine group (P<0.05). The expression of MST1 and Bax was higher in the taurine group than in the positive control group and the blank group (P<0.05), the expression of MST1 and Bax increased with increasing concentration in the taurine group (P<0.05). The expression of YAP and Bcl-2 was lower in the taurine group than in the positive control group and the blank group (P<0.05), the expression of YAP and Bcl-2 decreased with increasing concentration in the taurine group (P<0.05).Conclusions: Taurine promoted apoptosis and inhibited proliferation of prostate cancer cells, and its mechanism of action may be related to the MST1/Hippo signaling pathway in a dose-dependent manner.

Рак предстательной железы -одно из самых частых онкологических заболеваний у мужчин. Представления о роли основного мужского гормона -тестостерона -в генезе карциномы предстательной железы кардинально менялись за последние два с лишним века. Цель исследования -проанализировать эволюцию взглядов на взаимосвязь тестостерона и рака предстательной железы и оценить ее влияние на организацию медицинской помощи. Материалы и методы. Поиск публикаций осуществлялся в Национальной медицинской библиотеке США и Центральной научной медицинской библиотеке Российской Федерации по ключевым словам: рак предстательной железы, тестостерон. Учитывались только оригинальные публикации и метаанализы. В итоге было отобрано 59 публикаций и проведен их описательный анализ. Результаты. Представлены сведения о сомнительности отрицательной роли тестостерона в канцерогенезе рака предстательной железы ввиду малого числа наблюдений и многочисленных методологических дефектов проведенных исследований. Обозначена доказательность теории андрогенного насыщения при карциономе предстательной железы, которая основана на работах с достаточным количеством наблюдений. Описаны исследования, подтверждающие отсутствие канцерогенного влияния андрогензаместительной терапии у здоровых индивидов и прогрессии опухолевого процесса на фоне назначения тестостерона после лечения онкологического процесса. Более того, приведены сведения о применении указанного гормона при лечении кастрационно-резистентной формы карциномы предстательной железы. Заключение. С течением времени появляется все больше данных, которые подтверждают протективную роль тестостерона в отношении развития и даже при лечении рака предстательной железы. В связи с этим возникает перспектива глобальной трансформации организации медицинской помощи в вопросах тактики ведения больных карциномой предстательной железы, а также в целом мужчин старших возрастных групп. Очевидной остается необходимость проведения новых более качественных исследований по указанному вопросу.

Introduction Gender dysphoria is the discrepancy between biological sex and gender identity. This can be debilitating for transgender populations, including transgender men (TM), individuals who were assigned female at birth but who identify as men, that can benefit from hormonal therapy with testosterone products to address gender dysphoria. Methods We aim to summarize the efficacy, safety profile, and outcomes of the different testosterone replacement treatment (TRT) in the TM population. A search of the published literature regarding the various FDA‐approved TRT was performed in PubMed, Web of Science and Cochrane Library from 2007 to date. Results We complied two groups of TRT based on route of administration including the conventional testosterone therapies (intramuscular and subcutaneous injectables, and transdermal gels) and newer testosterone therapies (oral, buccal, and nasal gels). For the conventional testosterone therapies, we identified nine studies discussed conventional TRT in TM population including one randomized trial, four prospective studies, one retrospective study and three reviews. For newer testosterone therapies, we identified three studies discussed newer TRT in TM population including one prospective study and two reviews. Articles were then compiled and analyzed. Albeit majority of TRT data stemming from conventional TRT, there appear to be an overwhelmingly safety and efficacy profile in TM population translated with increased free testosterone levels comparable to male range, menses cessation, anxiety/depression decline and improved quality of life. Conclusion Testosterone therapy can be impactful for TM population with improved safety, efficiency, quality of life and function. With the rise of the newer FDA‐approved TRT, randomized studies are warranted to determine its safety and efficacity in this TM population.
