Abstract: Tendons regenerate poorly due to a dense extracellular matrix and low cellularity. Cellular therapies aim to improve tendon repair using mesenchymal stem cells and tenocytes; however, a current limitation is the low proliferative potential of tenocytes in cases of severe trauma. The purpose of this study was to develop a method useful in veterinary medicine to improve the differentiation of Peripheral Blood equine mesenchymal stem cells (PB-MSCs) into tenocytes. PB-MSCs were used to study the effects of the ad… Show more

“…All batches between P6 and P8 were seeded in T75 flasks and tenogenically induced by supplementing tendogenic growth factors to the expansion medium until 80% confluency was reached and tenogenic differentiation markers were upregulated ( 19 , 21 , 22 ). A higher passage was used to allow upscaling in future cell manufacturing processes, which is considered to be possible thanks to the phenotypic stability of PB-MSCs from P5 up to P10 ( 21 , 23 , 24 ). After trypsinization, all the cells were resuspended in 1 ml DMEM low glucose (Life Technologies) with 10% of dimethyl sulfoxide (Sigma).…”

“…32 No difference was observed in the amounts of PCNA, a marker of cell proliferation, although some studies demonstrate the performance of ASCs in increased cell proliferation in tendon repair. 8,12,33 A study of Dex et al 34 also demonstrates the role of Tnmd in the maturation of collagen fibres. Therefore, although no significant difference was detected in the protein quantification of Tnmd, there was a trend towards a higher amount of Tnmd in the L, SCL, SC and T groups, respectively, corroborating the higher degree of organization of the collagen fibres.…”

“…6 The photobiostimulation promoted by the low-level laser (LLL) has attracted the attention of researchers in recent years, already being used in clinical practice for the treatment of tendinous and post-operative injuries, accelerating the repair process, with the improvement of the tissue reorganization. 9 Although some in vitro studies demonstrate the efficiency of LLL treatment combined with MSCs, 8,[10][11][12] the literature is scarce considering the association of both therapies in vivo. 8 Thus, the combination of LLL and ASCs in vivo could induce a greater proliferation of these cells in the injured region, considering small the percentage of ASCs that survive at the injury site after application.…”
