Telomere length, chromatin structure and chromosome fusigenic potential

Abstract: Telomeres are specialized structures at chromosome ends that are thought to function as buffers against chromosome fusion. Several studies suggest that telomere shortening may render chromosomes fusigenic. We used a novel quantitative fluorescence in situ hybridization procedure to estimate telomere length in individual mammalian chromosomes, and G-banding and chromosome painting techniques to determine chromosome fusigenic potential. All analysed Chinese hamster and mouse cell lines exhibited shorter telomere… Show more

“…The fusion of chromosome fragments as revealed by ITS appearance is frequently observed in irradiated mammalian somatic cells (Slijepcevic et al, 1997). In the present study, the chromosome fragments missing centromeres might have to undergo a process of DNA incorporation into intact chromosomes.…”

“…Numerous studies report increased incidence of anaphase bridges when TL and/or integrity is compromised (33)(34)(35)(36); however, to our knowledge, evidence of telomeric DNA amplification through this mechanism has not been reported. DC formation is a known initiator of BFB cycling, gene amplification, and altered gene dosage (34,36).…”

“…Los telómeros alargados que carecen de ADN normal o marcas de metilación de histonas también resultan en un aumento de recombinaciones teloméricas (20). Aunque es la estructura de la cromatina la determinante de la recombinación, los telómeros más largos experimentan recombinaciones con más frecuencia que los más cortos (20,21).…”
