Telocytes are a special type of interstitial cells characterized by distinctive cellular extensions with alternating thin segments (podomers) and dilations (podoms). Telocytes establish contact with various cells and structures, but their role in the regulation of the function of many cell types is still obscure. The aim of the current study was to investigate the morphology, histochemistry, and immunohistochemistry of telocytes, and their distribution, organization, and morphometric measurements in different layers of the adult bovine uterine tube. Telocytes showed positive immunostaining for CD117, S-100 protein, vimentin, desmin, α-smooth muscle actin, tubulin, laminin, estrogen receptor-α, and progesterone receptor. They were organized in different types of sheaths: subepithelial, inner/outer perimuscular, and intramuscular sheaths. Telocytes were scattered in the lamina propria, in the muscular layer, and the serosa. According to their size, they were grouped into different types of telocytes: small, large, and giant telocytes. Small telocytes were the most common type and located in all layers; large telocytes were observed in the epithelium, lamina propria, and inner/outer perimuscular and intramuscular sheaths, and giant telocytes were found in the external layer of the outer perimuscular sheath. Telocytes were connected by thin and thick telopodes (fenestrated membranes). Fenestrated membranes enabled connections between telocytes along the entire muscular wall of the uterine tube. Telocytes established an extensive biological network of different types of cells and structures, including epithelial, muscular, and mast cells, blood vessels, glomus, and nerve fibers. We hypothesize that telocytes help to organize the functional coordination between different types of cells in the uterine tube.

Telocytes (TCs) are a controversial cell type characterized by the presence of a particular kind of prolongations, known as telopodes, which are long, thin, and moniliform. A number of attempts has been made to establish the molecular phenotype of cardiac TCs (i.e., expression of c-kit, CD34, vimentin, PDGRFα, PDGRFβ, etc.). We designed an immunohistochemical study involving cardiac tissue samples obtained from 10 cadavers with the aim of determining whether there are TC-like interstitial cells that populate the interstitial space other than the mural microvascular cells. We applied the markers for CD31, CD34, PDGRFα, CD117/c-kit, and α-smooth muscle actin (α-SMA). We found that, in relation to two-dimensional cuts, the endothelial tubes could be misidentified as TC-like cells, the difference being the positive identification of endothelial lumina. Moreover, we found that cardiac pericytes express PDGRFα, CD117/c-kit, and α-SMA, and that they could also be misidentified as TCs when using light microscopy. We reviewed the respective values of the previously identified markers for achieving a clear-cut identification of cardiac TCs, highlighting the critical lack of specificity.

Endothelial expression of c-kit and CD68 in dental follicles of human impacted third molars

Periodontal tissue remnants of odontogenesis constitute the dental follicle (DF) which is actually considered a stem niche in adults. However, potentialities of local endothelia within this niche seem overlooked. We thus aimed at testing the endothelial cells expression of c-kit, the progenitor cells marker, and CD68, commonly regarded as a monocyte/macrophage marker, in human DFs. We performed an immunohistochemical study using these two markers which were applied on samples collected from ten adult patients. The markers were positively expressed in endothelial cells, as well as in spindle-shaped stromal cells of the DF.The origin of DF stem or progenitor cells needs reviewing in the light of these findings, as endothelium could be a donor site for niche inhabitants.
