Heat shock proteins (Hsps) are highly conserved molecular chaperones that are ubiquitously expressed in all species to aid the solubilization of misfolded proteins, protein degradation, and transport. Elevated levels of Hsp70 have been found in the sputum, serum, and bronchoalveolar lavage (BAL) fluid of asthma patients and are known to correlate with disease severity. However, the function of Hsp70 in allergic airway inflammation has remained largely unknown. This study aimed to determine the role of Hsp70 in airway inflammation and remodeling using a mouse model of allergic airway inflammation. WT and Hsp70 double-knockout (Hsp70.1/.3 ؊/؊) mice were sensitized and challenged intratracheally with Schistosoma mansoni soluble egg antigens (SEAs) to induce robust Th2 responses and airway inflammation in the lungs. The lack of Hsp70 resulted in a significant reduction in airway inflammation, goblet cell hyperplasia, and Th2 cytokine production, including IL-4, IL-5, and IL-13. An analysis of the BAL fluid suggested that Hsp70 is critically required for eosinophilic infiltration, collagen accumulation, and Th2 cytokine production in allergic airways. Furthermore, our bone marrow (BM) transfer studies show that SEA-induced airway inflammation, goblet cell hyperplasia, and Th2 cytokine production were attenuated in WT mice that were reconstituted with Hsp70-deficient BM, but these effects were not attenuated in Hsp70-deficient mice that were reconstituted with WT BM. Together, these studies identify a pathogenic role for Hsp70 in hematopoietic cells during allergic airway inflammation; this illustrates the potential utility of targeting Hsp70 to alleviate allergen-induced Th2 cytokines, goblet cell hyperplasia, and airway inflammation.

Purpose: Exercise modifies airway immune responses and susceptibility to infection. We investigated the effects of exercise on two HSPs (heat shock proteins), quantifying expression levels in nasal mucosa of both professional competitive athletes and non-athletes for comparison. Method: We used western blot technique to determine expression levels of HSPs in primary human nasal epithelial cells (HNECs). Nasal lavage (NAL) fluids were collected from 12 male professional volley ball players and 6 healthy males pre-submaximal exercise (running for 30 min at 70-80% of maximal heart rate) and post-submaximal exercise. Expression levels of HSP27, HSP70, Interleukin (IL)-8, and Tumor necrosis factor (TNF)-α in NAL fluids were quantified by enzyme-linked immunosorbent assay (ELISA), and difference of the level between pre-submaximal exercise and post-submaximal exercise was statistically analyzed. Antibacterial assay using Staphylococcus aureus was performed to assess the immunological role of HSPs in NAL fluids. Results:. In non-athlete controls, HSP27, HSP70, and IL-8 were unchanged after exercise. In the professional athletes, HSP70 expression was declined significantly (p<0.05), but HSP27 was not significantly changed. IL-8 and TNF-α did not show significant difference, either. By antibacterial assay, it was found that the number of active bacterial populations were influenced by the presence or absence of HSP27 and HSP70 in NAL fluids. Conclusion: HSP27 and HSP70 were present in NAL fluids of enrolled subjects, and the effect of exercise on the level HSPs was different between professional athletic competitors and non-athletes. As the number of active bacterial population was influenced by the presence or absence of nasal HSP27 and HSP70, we suggest that HSP27 and HSP70 may play immunological function in NAL fluids.

Aim. To determine the content of heat shock proteins with molecular weight 70 kDa (HSP 70) and antibo dies to them in blood and nasal secretion in patients with allergic rhinites and infectious rhinosinusites of different etiology. Materials and methods. The paper presents the results of examination of 10 patients with allergic rhinitis and 30 patients, infected with rhinosinusites(the age range 25–35 years). The patients with infectious rhinosinusites were divided into 3 groups according to nosologic form of disease. The control group included 10 practically healthy persons in the ratio, comparable by their gender and age with sick persons. Results.The analysis showed that in the nasal secretion of all patients, HSP 70 level significantly raised compared to the control. Maximum values were registered in patients with bacterial rhinosinusitis and were higher than in patients with viral and fungous ones by 1.9 times (p = 0.015) and 2.9 times (p = 0.001), respectively. In blood serum, HSP 70 concentration compared with the control increased in patients with allergic rhinitis and bacterial rhinosinusitis by 103.67 % (p = 0.015) and 32.11 % (p = 0.049), respectively; these values in the last two groups exceeded the latter in patients with fungous RS by 2.37 times (p = 0.01) and by 1.54 times (p = 0.035). Conclusions. It was detected that in the group of patients with allergic rhinitis and chronic bacterial rhinosinusitis in the nasal secretion and blood serum, HSP 70 values were the highest. In the nasal secretion, HSP 70 level was higher than in blood. The amount of autoantibodies to HSP 70 in blood grew in allergic rhinitis, fungous and viral forms of rhinosinusites that reflects the immunological effect of chaperone proteins.
