Biological Role of Gellan Gum in Improving Scaffold Drug Delivery, Cell Adhesion Properties for Tissue Engineering Applications

Over the past few decades, gellan gum (GG) has attracted substantial research interest in several fields including biomedical and clinical applications. The GG has highly versatile properties like easy bio-fabrication, tunable mechanical, cell adhesion, biocompatibility, biodegradability, drug delivery, and is easy to functionalize. These properties have put forth GG as a promising material in tissue engineering and regenerative medicine fields. Nevertheless, GG alone has poor mechanical strength, stability, and a high gelling temperature in physiological conditions. However, GG physiochemical properties can be enhanced by blending them with other polymers like chitosan, agar, sodium alginate, starch, cellulose, pullulan, polyvinyl chloride, xanthan gum, and other nanomaterials, like gold, silver, or composites. In this review article, we discuss the comprehensive overview and different strategies for the preparation of GG based biomaterial, hydrogels, and scaffolds for drug delivery, wound healing, antimicrobial activity, and cell adhesion. In addition, we have given special attention to tissue engineering applications of GG, which can be combined with another natural, synthetic polymers and nanoparticles, and other composites materials. Overall, this review article clearly presents a summary of the recent advances in research studies on GG for different biomedical applications.

Objective: The purpose of the present work was to enhance antimicrobial activity of Suppress release Amoxicillin Camphor complexed Mucoadhesive tablets by Statistical Experimental Design using Sigma tech software version 3.1. Amoxicillin has short half-life (1 h) which requires frequent administration of the conventional amoxicillin tablets and is mainly used in the eradication of Helicobacter pylori that resides in the stomach. Methods: Amoxicillin-Sodium cholate (ASC) and camphor complexes (ACC) were prepared to enhance antimicrobial activity. Results: TLC and FT-IR studies confirmed the formation of drug complex. Zone of inhibition by agar well diffusion method of ACC showed greater inhibition than ASC, solubility of ACC was enhanced by re-crystallization technique. Hence ACC needle shaped re-crystallized was used to sustained release tablets using mucoadhesive polymers. Chitosan, HPMC K-15 and Ethyl cellulose (EC) were selected as independent variables and ex vivo mucoadhesion time, % drug release at 24 h and t 50 % (time to release 50 % drug) were selected as dependent variables. DSC studies indicated drug and excipients were compatible. Swelling studies and scanning electron microscopic analysis confirmed the drug release mechanism from sustained release tablets. In-vitro release studies and ex vivo studies of amoxicillin confirmed the sustained drug release profile with first order release kinetics better mucoadhesion and enhanced antimicrobial activity. The optimized formulation was found to be stable at accelerated storage conditions for 3 months. Conclusion: The results demonstrated the effectiveness of the proposed statistical design for optimization of ACC sustained release tablets.

Investigation of the effects of viscoelasticity and hydration kinetic on the drug release behavior from the poly (sodium acrylate)-grafted-gellan matrix (PSAc-g-GG) was the main objective of this study. At first, poly (acrylic acid)-grafted-gellan was treated with 0.05M NaOH to obtain poly (sodium acrylate)-grafted-gellan followed by its purification and subsequent lyophilicity study and viscoelastic study on PSAc-g-GG with different degree of grafting. The study revealed that the degree of grafting greatly affects the viscoelastic and rheologic characteristics of the copolymer, which further affect the drug release profile from the polymeric matrix. The copolymer with highest grafting (626.3%) exhibited much higher starting % strain (17.79%), stress (53.7 Pa) for structural breakdown at Gꞌ = Gꞌꞌ (214.4 Pa), higher storage modulus (G’), much greater values of complex viscosity (11.5 Pa.s) and cross-over point (Gꞌ = Gꞌꞌ =271.65 Pa) compared to that of the batch of copolymer with lower grafting. The water uptake index (%WE) was found to be directly proportional to the percentage grafting (%G), whereas the batches with higher grafting revealed lower initial swelling rate representing its inversely proportional relation to %grafting in case of 0.1N HCl acid. Equilibrium swelling and hydration were also found to be proportional to % grafting. The similar effect was observed in phosphate buffer solution (pH 6.8) with an exception that the degree of the swelling parameters obtained from phosphate buffer was very much greater compared to that found in 0.1N HCl. PSAc-g-GG exhibited extended drug release over a period of 10 hours with the drug release mechanism based on Case-1 Fickian diffusion or square root of time kinetic. The study also exhibited the usefulness of viscoelastic and swelling study in order to identify the effects of the degree of grafting on the drug release.
