Survey of commercial Rhodiola products revealed species diversity and potential safety issues

Abstract: The adulteration of herbal products is a threat to consumer safety. Here we surveyed the species composition of commercial Rhodiola products using DNA barcoding as a supervisory method. A Rhodiola dietary supplement DNA barcode database was successfully constructed using 82 voucher samples from 10 Rhodiola species. Based on the DNA barcoding standard operating procedure (SOP), we used this database to identify 100 Rhodiolae Crenulatae Radix et Rhizoma decoction piece samples that were purchased from drug store… Show more

“…Compared with other candidate DNA barcodes, namely, psbA-trnH, matK, rbcL, rpoC1, ycf5, and ITS, the ITS2 sequence represents the most suitable region for DNA barcoding applications to classify medicinal plant species [22]. Most medicinal plants such as Cornus officinalis, Menispermum dauricum, Rhodiola crenulata, and Selaginella tamariscina could be identified effectively based on ITS2 barcode [19,20,35,39]. In the present study, the ITS2 region was used to distinguish Shi-Liang tea species from closely related species.…”

“…DNA markers have become convenient tools for species identification and molecular systematics as a result of the advances in molecular technology [34][35][36]. However, ITS/ITS2 regions as universal barcodes present several problems and limitations, such as ITS/ITS2 showed poor PCR success in non-angiosperms [36].…”

“…The recently published results, along with the growing number of reports on the authenticity and quality control of R. rosea products (Xin, 2015;Booker et al 2015) are increasingly justifying the importance of in vitro systems optimization for reliable and homogenous plant material production. “…The use of authentic medicinal plant species is a fundamental requirement in herbal medicine [12]. Adulteration, substitution, and mislabelling may cause potential harm to patients [13][14][15][16][17]. Along with safety and toxicity, there is a need to develop methods of profiling medicinal plants for the purpose of authentication.…”
