Sulfonate-functionalized tetraphenylethylenes for selective detection and wash-free imaging of Gram-positive bacteria (Staphylococcus aureus)

Abstract: Two sulfonate functionalized tetraphenylethylene (TPE) derivatives were synthesized and used as probes for the detection and imaging of Gram-positive bacteria (e.g. Staphylococcus aureus).

“…In general, AIE‐sulfonates are non‐emissive in aqueous solution but fluoresce strongly in the aggregated state, either by the formation of water‐dispersible aggregates or by adhering onto solid surfaces (e. g. cell‐wall of bacteria) (Figure 2a) [35,38,44,45] . The aqueous solubility of AIE‐sulfonates increases with the increasing number of −SO 3 − functionalities in the molecule.…”

“…designed a tetraphenylethylene (TPE) derivative with two sulfonate groups ( Figure ). [ 96 ] The negatively charged AIE probe TPE‐di‐sulfonate showed strong electrostatic interaction with the amino groups of lipoteichoic acid, which restricted the intramolecular motion of TPE‐di‐sulfonate, thus exhibiting 43 folds increase of fluorescent intensity upon incubating with Gram‐positive Staphylococcus aureus or Bacillus subtilis . However, negligible fluorescence was observed upon TPE‐di‐sulfonate was incubated with Gram‐negative E. coli or Pseudomonas aeruginosa .…”
