Stromal Cells Are the Main Plasminogen Activator Inhibitor-1-Producing Cells in Human Fat

Abstract: Abstract-Elevated plasma plasminogen activator inhibitor (PAI)-1 observed during insulin resistance has been connected with an excessive PAI-1 adipose tissue secretion mainly by visceral fat. Our aim was to compare the localization of PAI-1 in human visceral and subcutaneous fats. PAI-1 secretion was also investigated in vitro during human adipocyte differentiation. PAI-1 antigen and mRNA were localized in the stromal area of the tissue and were also present in a few CD14-positive monocytes, in direct contact … Show more

“…2A, adipose tissue expression of TSP1 and PAI-1 mRNAs associated positively (r ϭ 0.48, P Ͻ 0.0001, n ϭ 80). Interestingly, PAI-1 gene expression was higher in the SVF compared with the adipocyte fraction (1.76 Ϯ 0.32 and 0.49 Ϯ 0.14, respectively), consistent with previous reports (32), that is reciprocal to the expression of the TSP1 gene (shown in Table 1), which predominates in the adipocyte fraction. A positive correlation was also observed on examining the association of adipose tissue TSP1 mRNA and circulating PAI-1 levels (r ϭ 0.36, P Ͻ 0.02, n ϭ 41, Fig.…”

“…By contrast, PAI-1 was expressed predominantly in the SVF (Fig. 1c), consistent with the fact that it is mainly expressed by adipose tissue macrophages [24]; leptin was expressed 1,000-fold higher in the AF than in the SVF (Fig. 1d) and adiponectin 400-fold higher (Fig.…”

“…For stromal tissue close to adipocytes, the stained stromal area was divided by the number of adipocytes counted in the examined field (labeled stromal surface/ adipocyte). Antibody against leptin was used as positive control of adipocyte staining and immunohistochemistry was performed as already described (22).…”
