Stratum corneum lipid abnormalities in ichthyosis. Detection by a new lipid microanalytical method

Abstract: Although the biochemical diagnosis of the ichthyoses is still in its infancy, the two recessively inherited types, recessive X-linked ichthyosis (RXLI) and nonbullous congenital ichthyosiform erythroderma (CIE), are accompanied by stratum corneum lipid abnormalities. However, in RXLI, cholesterol sulfate accumulates; in CIE, massive quantities of n-alkanes accumulate. The diagnosis of these disorders has required large quantities of scale for sequential, quantitative thin-layer chromatography (TLC). In this st… Show more

“…4) (23). t Results of stratum granulosum vs. spinosum/basale in normal epidermis and of stratum corneum vs. stratum granulosum or stratum spinosum/basale in RXLI, are statistically significant (P < 0.01, < 0.05, respectively).…”

Exaggerated anticipatory anxiety is common in social anxiety disorder (SAD). Neuroimaging studies have revealed altered neural activity in response to social stimuli in SAD, but fewer studies have examined neural activity during anticipation of feared social stimuli in SAD. The current study examined the time course and magnitude of activity in threat processing brain regions during speech anticipation in socially anxious individuals and healthy controls (HC). Method Participants (SAD n = 58; HC n = 16) underwent functional magnetic resonance imaging (fMRI) during which they completed a 90s control anticipation task and 90s speech anticipation task.

“…Lipids were extracted as previously described (Williams et al, 1985a). Quantification of lipids was performed by two methods: thin layer chromatography (TLC) followed by lipid extraction and weighing (Elias et al, 1979) and quartz rod microchromatography-flame ionization (Iatroscan ~) (Brown et al, 1984). The distribution of neutral lipid classes and determination of total polar lipids was determined using the Iatroscan R. This method offers the advantages of analysing complex tipids from small amounts of starting material and compared favourably with other methods of analysis (Rao et al, 1985;Kramer et al, 1985).…”

“…For quantitation of the small amounts of lipid in these cultures, we employed quartz rod microchromatography (Ackman, 1981;Brown et al, 1984). Lipids were dried down under nitrogen gas and resuspended in warm chloroform:methanol (2:1, vols) to a final lipid concentration of about 20 pg/pL.…”
