Naturally competent bacteria acquire DNA from their surroundings to survive in nutrient-poor environments and incorporate DNA into their genomes as new genes for improved survival. The secretin HofQ from the oral pathogen Aggregatibacter actinomycetemcomitans has been associated with DNA uptake. Cytokine sequestering is a potential virulence mechanism in various bacteria and may modulate both host defense and bacterial physiology. The objective of this study was to elucidate a possible connection between natural competence and cytokine uptake in A. actinomycetemcomitans. The extramembranous domain of HofQ (emHofQ) was shown to interact with various cytokines, of which IL-8 exhibited the strongest interaction. The dissociation constant between emHofQ and IL-8 was 43 nM in static settings and 2.4 μM in dynamic settings. The moderate binding affinity is consistent with the hypothesis that emHofQ recognizes cytokines before transporting them into the cells. The interaction site was identified via crosslinking and mutational analysis. By structural comparison, relateda type I KH domain with a similar interaction site was detected in the Neisseria meningitidis secretin PilQ, which has been shown to participate in IL-8 uptake. Deletion of hofQ from the A. actinomycetemcomitans genome decreased the overall biofilm formation of this organism, abolished the response to cytokines, i.e., decreased eDNA levels in the presence of cytokines, and increased the susceptibility of the biofilm to tested β-lactams. Moreover, we showed that recombinant IL-8 interacted with DNA. These results can be used in further studies on the specific role of cytokine uptake in bacterial virulence without interfering with natural-competence-related DNA uptake.

The Yersinia pestis protein Caf1M is a typical representative of a subfamily of periplasmic molecular chaperones with characteristic structural and functional features, one of which is the location of two conserved cysteine residues close to the putative binding pocket. We show that these residues form a disulphide bond, the reduction and alkylation of which significantly increases the dissociation constant of the Caf1M-Caf1 (where Caf 1 is a polypeptide subunit of the capsule) complex [from a K d of (4.77p0.50)i10 −* M for the intact protein to one of (3.68p0.68)i10 −) M for the modified protein]. The importance of the disulphide bond for the formation of functional Caf1M in i o was demonstrated using an Escherichia coli dsbA mutant carrying the Y. pestis f1 operon. Резюме. Чума -зоонозное заболевание, вызванное грамотрицательной бактерией Yersinia pestis, которая, как правило, передается человеку от зараженных грызунов через укус инфицированной блохи. Данный микро-организм убил больше людей, чем все войны в истории человечества. Циркуляция Y. pestis в природных оча-гах чумы обеспечивается целым рядом факторов патогенности различной функциональной направленности. В обзоре рассматривается один из них -капсульный антиген чумного микроба (F1 или Caf1). Описаны исто-рия открытия, генетический контроль, условия биосинтеза, выделение, очистка и физико-химические свой-ства капсульного антигена, а также обсуждается его вклад в патогенез чумы и его использование в качестве основного компонента чумных вакцин. Визуально аморфная при световой микроскопии капсула Y. pestis при электронной микроскопии высокого разрешения представляет собой структуру, сформированную из отдель-ных фимбриеподобных тяжей длиной до 200 нм, расходящихся в разные стороны от поверхности бактерии. При температуре 37°С образуется в 800-1000 раз больше капсульного антигена, чем при 28°С. Гены, кодирую-щие белок Caf1 с молекулярной массой субъединицы 17,6 kDa, состоящей из 170 аминокислот, локализованы в caf1 опероне на плазмиде pFra. На основе анализа первичной нуклеотидной последовательности caf1 оперо-на было установлено его филогенетическое родство с кластерами генов, кодирующих секретируемые с помо-щью шаперон/ашерных систем пилевые адгезины не только других энтеробактерий, но и еще шесть адгезинов чумного микроба. Размножение клеток Y. pestis внутри макрофагов является обязательным этапом патогенеза чумы, а вирулентность чумного микроба коррелирует не с устойчивостью к захвату фагоцитами, а со способ-ностью выживать и размножаться в фаголизосомах фагоцитарных клеток за счет подавления антибактери-альных функций фагоцитов. Капсула, образованная из агрегатов Caf1, защищает клетки Y. pestis от захвата интактными фагоцитами хозяина, препятствует инициации альтернативного пути активации комплемента. Молекулярный ашер Caf1A, ответственный за закрепление капсульного антигена на поверхнос ти бактери-альной клетки, имеет высокий аффинитет к человеческому интерлейкину 1β. Caf1 может вступать в конку-ренцию с интерлейкинами 1α, 1β и 1ra за связывание с рецепторами на лимфоидных клетках, препятствуя развитию адекватного иммунного ответа. Иммунодиагностика чумы построена на выявлении Caf1 или анти-Caf1-антител, так как этот антиген Y. pestis видоспецифичен. Покрывающий поверхность микробных клеток капсульный антиген -важнейший составной элемент всех современных чумных вакцин. Показана его ве-дущая роль в создании напряженного иммунитета у белых мышей, крыс, приматов и человека. Однако в ор-ганизме иммунного животного могут накапливаться бескапсульные (Caf1 -) варианты Y. pestis, сохранившие вирулентность на уровне исходных штаммов «дикого типа».
