Solid‐state synthesis of cyclo LD‐diphenylalanine: A chiral phase built from achiral subunits

Abstract: The solid-state structure of LL/DD or LD/DL diphenylalanine diluted in KBr pellets is studied by infrared (IR) absorption and vibrational circular dichroism (VCD) spectroscopy. The structure depends on the absolute configuration of the residues. The natural LL diphenylalanine exists as a mixture of neutral and zwitterionic structures, depending on the humidity of the sample, while mostly the zwitterion is observed for LD diphenylalanine whatever the experimental conditions. The system undergoes spontaneous cyc… Show more

“…The absolute configuration of the residues indeed influences the structure of cyclic dipeptides. [36][37][38][39][40][41] Page 4 of 35 Physical Chemistry Chemical Physics Cyclic dipeptides differ from linear ones in several aspects. First, the steric bias due to the ring imposes the non-usual cis conformation of the peptide bonds.…”

“…The appropriate reaction temperature depends on the nature of the peptide sequence. For example, Gly-Gly needs 230 °C, while Phe-Phe 147 °C [ 90 ] or 125 °C [ 91 ], suggesting a correlation between the reaction minimum temperature and the amino acid side-chain steric hindrance. Cyclization indeed requires molecular mobility, which is governed by the nature of the side chains in the solid state.…”

“…The stereochemical effects of the self‐assembly of tripeptide Phe‐Phe‐Phe were revealed by solid‐state VCD 26 . The solid‐state structure of cyclo ld ‐diphenylalanine 27 and the helicity of Ala‐Ala dipeptides with a perfluoroalkyl chain at the N‐terminal were studied using VCD 28 . Representative VCD couplet patterns for the amide I and amide II vibrational modes of polypeptides have been presented in some previous studies 29–31 .…”
