The aim of this study was to determine the effects of coated calcium butyrate (CCB) on calf performance and some blood parameters. Sixteen female Holstein calves with a mean age of 3 ± 1 d were divided into two equal groups and fed a milk replacer supplemented with 3 g of coated calcium butyrate (CCB)/day or with no coated calcium butyrate (NCB). The calves had free access to solid feed and water. Body weight was measured at days 3, 12, 24, 36 and 48 (weaning day). Respiratory rate and rectal temperature (within first 4 weeks of life), feed intake and faecal score (during the whole experimental period) were recorded daily. Rumen fluid was taken for volatile fatty acid (VFA) determination on days 33 and 48, and structural growth (rump height, withers height and hip width) was recorded on days 3, 24 and 48. Blood samples were collected on days 1, 12, 24, 36 and 48. Feed intake, average daily gain, feed conversion ratio and structural growth of calves were improved by CCB supplementation. There were no significant differences on rectal temperature, respiratory rate and faecal score between the treatments. Rumen concentration of total VFAs increased and the VFA profile was affected on days 33 and 48 by CCB supplementation. The supplementation of milk replacer of calves with coated calcium butyrate significantly increased serum concentrations of glucose, insulin and β-hydroxybutyrate. Serum concentration of cortisol in the CCB calves was numerically lower than in NCB. In conclusion, supplementation of calves' milk replacer with the coated calcium butyrate could improve calf performance. ________________________________________________________________________________

Background/Aims: Butyric acid plays an important role in maintaining intestinal health. Butyric acid has received special attention as a short-chain fatty acid, but its role in protecting the intestinal barrier is poorly characterized. Butyric acid not only provides energy for epithelial cells but also acts as a histone deacetylase inhibitor; it is also a natural ligand for G protein-coupled receptor 109A (GPR109A). A GPR109A analog was expressed in Sus scrofa and mediated the anti-inflammatory effects of beta-hydroxybutyric acid. This study investigated the effects of butyrate on growth performance, diarrhea symptoms, and tight junction protein levels in 21-day-old weaned piglets. We also studied the mechanism by which butyric acid regulates intestinal permeability. Methods: Twenty-four piglets that had been weaned at an age of 21 days were divided randomly into 2 equal groups: basal diet group and sodium butyrate + basal diet group. Diarrhea rate, growth performance during 3 weeks of feeding on these diets were observed, the lactulose-mannitol ratio in urine were detected by High Performance Liquid Chromatography, the expression levels of tight junction proteins in the intestinal tract and related signaling molecules, such as GPR109A and Akt, in the colon were examined by quantitative real-time PCR or western blot analyses on day 21. Caco-2 cells were used as a colon cell model and cultured with or without sodium butyrate to assess the expression of tight junction proteins and the activation of related signaling molecules. GPR109A-short hairpin RNA (shRNA) and specific antagonists of Akt and ERK1/2 were used as signaling pathway inhibitors to elucidate the mechanism by which butyric acid regulates the expression of tight junction proteins and the colonic epithelial barrier. Results: The sodium butyrate diet alleviated diarrhea symptoms and decreased intestinal permeability without affecting the growth of early weaned piglets. The expression levels of the tight junction proteins Claudin-3, Occludin, and zonula occludens 1 were up-regulated by sodium butyrate in the colon and Caco-2 cells. GPR109A knockdown using shRNA or blockade of the Akt signaling pathway in Caco-2 cells suppressed sodium butyrate-induced Claudin-3 expression. Conclusions: Sodium butyrate acts on the Akt signaling pathway to facilitate Claudin-3 expression in the colon in a GPR109A-dependent manner.

A total of 96 pigs (Duroc×Landrace×Yorkshire) weaned at day 21 with an average initial body weight of 6.68 kg were used to investigate the effects of sodium butyrate on the growth performance, intestinal microfl ora and morphology of weanling pigs. The pigs were allocated to three dietary treatments in a randomized complete block design, each treatment was replicated four times with eight pigs per replicate and the trial lasted for 30 days. The dietary treatments were: 1. basal diet, 2. basal diet + 500 mg/kg sodium butyrate, 3. basal diet + 1000 mg/kg sodium butyrate. Diets were formulated according to requirements of NRC (1998) for 10-20 kg pigs. All diets were fed as mash, and no antibiotics was included. The results showed that supplementation with 1000 mg/kg sodium butyrate improved (P<0.05) growth performance, reduced (P<0.05) the total viable counts of intestinal Clostridium and Escherichia coli, decreased (P<0.05) tumor necrosis factor-α (TNF-α) and interleukin-6 (IL-6) levels in the serum, and increased (P<0.05) villus height and the villus height to crypt depth ratio at the small intestinal mucosa as compared with control. The results indicated that sodium butyrate is effective in enhancing the growth and intestinal morphology of weanling pigs.
