Abstract: Retrovirus virions carry a diploid genome associated with a large number of small viral finger protein molecules which are required for encapsidation. Our present results show that finger protein p12 of Rous sarcoma virus (RSV) and p10 of murine leukaemia virus (MuLV) positions replication primer tRNA on the replication initiation site (PBS) at the 5′ end of the RNA genome. An RSV mutant with a Val‐Pro insertion in the finger motif of p12 is able to partially encapsidate genomic RNA but is not infectious becau… Show more

“…It has been demonstrated by several groups that nucleocapsid protein does not unwind the secondary structure of tRNA in vitro, and the protein only has very subtle tertiary structural and helix destabilization effects on tRNA alone (44)(45)(46)(47). Similar observations have been made in RSV and MoMuLV, where nucleocapsid protein promotes the annealing of tRNA Trp and tRNA Pro to the PBSs of the RSV and MoMuLV genomic RNA, respectively (48).…”

“…We speculate that fundamental aspects of genomic RNA dimerization and tRNA annealing derive from the potent intrinsic nucleic acid strand renaturation activity of NC proteins reported here. The mechanistic details may well be different to some extent, because, for example, previous findings published by other laboratories suggest that NC-saturated viral RNA (assuming napp ranging from 8 to 15) does not appear to be required in genomic dimer formation or tRNA positioning in vitro (Prats et al, 1988). However, in most in vitro genomic RNA dimerization assays, there is some evidence in support of a rapid reaction that appears to require a critical concentration of NC or molar ratio of NC/RNA nucleotide, below which the reaction seems inefficient (cf.…”

“…The previously reported ability of NC proteins to facilitate the intermolecular base-pairing between replication primer tRNA and complementary RNA or DNA sequences (Prats et al, 1988) can be viewed as a specialized case of a general nucleic acid strand renaturation activity. Proteins that bind preferentially to ss rather than duplex DNA can, under certain solution conditions, exhibit the ability to catalyze the rate of renaturation of nucleic acid strands.…”

“…Despite their relative small size, retroviral NC proteins have a variety of other functions. For instance it has been reported that they promote the annnealing of the replication primer tRNA to the RNA genome (21). Also preliminary data indicate that RSV NC can direct dimerization of retroviral RNA, a process that could be relevant to reverse transcription, since it has been shown that RNA template switching occurs during the synthesis of proviral DNA (21,22).…”
