Single nucleotide polymorphisms in Plasmodium falciparum V type H+ pyrophosphatase gene (pfvp2) and their associations with pfcrt and pfmdr1 polymorphisms

Abstract: The VKP haplotype was dominant. Most pfvp2 405I, 582R and 711S SNPs were seen where CQ resistance was not highly prevalent at the time of blood sampling possibly due to greater genetic variation prior to the bottle neck event of spreading CQ resistance. The association between the pfvp2 VKP haplotype and pfcrt 76T, which may indicate that pfvp2 is involved in CQ resistance, should therefore be interpreted with caution.

“…Irrespective of how this isolated CVIET haplotype came to be in the study area it did not seem to have become widespread as only pfcrt CVMNK parasites were found in 50 samples collected 3 years later in the same area [40]. This observation is consistent with longitudinal surveillance of high in vivo efficacy and in vitro susceptibility to CQ in Bondi (CQ village) between 1976 and 1978 [29, 41].…”

“…The studies show that on the Thailand-Myanmar border, the amplification rate is greater, reaching 100% [84, 85]. In general, Southeast Asian countries had a greater prevalence of P. falciparum isolates with pfmdr1 amplification, while isolates from Southwest Asia showed no gene amplification [8690]. Taken together, these differences in gene amplification rates may be the result of the adoption of different treatment regimens for malaria by different countries over the years.…”

“…8,12 There are several possible explanations for these apparent contradictions. Deep genome sequencing of CQR versus CQS parasite strains and isolates has identified various genes with mutations or altered expression that are associated with the presence of mutant PfCRT 14,15 that might abrogate mutant PfCRT fitness costs. It is also possible that additional mutations within PfCRT itself may reduce the fitness burden associated with some isoforms.…”
