ABSTRACT. Bovine papillomavirus (BPV) is an oncogenic virus with mucous and epithelial tropism. Possible productive virus infection in other tissues, such as blood, has been hypothesized. In order to investigate this possibility, three samples of skin papillomas and blood were collected from bovines with BPV infection and five samples of peripheral blood and one sample of normal tissue were collected from a calf without BPV infection. Peripheral blood mononuclear cells (PBMCs) were isolated from whole blood and examined by reverse transcription-polymerase chain reaction, immunofluorescence, in situ hybridization, and electron microscopy. The tissue samples were examined for histopathological and (2015) immunohistochemical features. The skin papillomas showed the presence of DNA sequences of BPV-2, BPV-11, and a putative virus type. The blood samples showed DNA sequences of BPV-1, 2, and 4 simultaneously. Immunohistochemistry showed BPV L1 protein in both epithelium and stroma and BPV E2 protein in koilocytes. In situ hybridization confirmed the presence of BPV DNA in PBMCs and immunofluorescence showed nuclear labeling of E2 and L1 BPV proteins in PBMCs. The transcription analysis revealed transcripts of BPV-1 L1, BPV-2 L2, and BPV-4 E7 in blood and papilloma samples of BPV-infected cattle. The comet assay revealed high levels of host cell DNA damage upon BPV infection. Electron microscopy analysis of PBMCs identified the presence of particles in the cytoplasm that are consistent with papillomavirus in size and shape. The productive infection of PBMCs with BPV has been previously discussed and this study provides evidence indicating that PBMCs are a target of BPV.

SummaryIn animals and man, some forms of cutaneous papillomas are caused by papillomaviruses. Twelve different genotypes of bovine papillomavirus (BPV) have been identified. BPV-1 through BPV-12 are all strictly species-specific. BPV-1 and BPV-2 are associated with fibropapillomas in cattle; these tumors are formed by excessive proliferation of virus-infected dermal fibroblasts and epidermal keratinocytes. In the present study well known diagnostic procedures including histopathological, immunohistochemical and molecular methods were used for the diagnosis of bovine papillomavirus infection.Samples were taken randomly from five cattle with multiple cutaneous tumor formations for the diagnosis of bovine papillomavirus infection occurring commonly in herds.In the present study five cattle were examined for the presence of multiple cutaneous tumors. Cutaneous fibropapillomatosis were confirmed histopathologically and immunohistochemically. BPV-1 and BPV-2 which are etiological agents for the bovine cutaneous fibropapillomatosis were investigated by PCR. BPV serotyping was performed from five cattle all coming from three different farms. In conclusion papillomaviral DNA was detected by the PCR. The amplified long control region (LCR) DNA sequence was identical to that of BPV-1 and negative for the BPV-2 DNA. When considered economical impact of the disease associated with the abundance of the infection significance of protective measures and vaccine studies come forward. Sığırlarda BPV-1 ve BPV-2 ile enfekte dermal fibroblast ve epidermal keratonositlerin aşırı çoğalması sonucunda fibropapillomlar oluşmaktadır. Bu çalışmada sığırların deri tümörlerinde etiyolojik ajanlardan biri olan papilloma viruslarının rolü araştırılmıştır. Papillomların yaygın olarak bulunduğu sürülerde, BPV enfeksiyonunun teşhisi amacıyla; rasgele seçilen multiple deri tümörleri bulunan beş sığırdan örnekler alındı. Araştırmada papilloma enfeksiyonunun teşhisine yönelik olarak dünyada uygulanırlığı kabul edilen histopatolojik, immunohistokimyasal ve moleküler yöntemler kullanıldı. Histopatolojik ve immunohistokimyasal metotlarla deri fibropapillomatozisleri doğrulandıktan sonra, sığır deri fibropapillomatozislerine neden olan BPV-1 ve BPV-2 nin rolü, üç çiftlikten alınan beş sığır örneğinde moleküler olarak PZR (polimeraz zincir reaksiyonu) yöntemi ile araştırıldı. Sonuçta PZR metodu ile BPV-1'in DNA zincirindeki kontrol bölgesi tespit edilmesine rağmen BPV-2 negatif bulundu. Etiyolojik nedeni ortaya konan bu araştırma ile enfeksiyonun yaygınlığına bağlı ekonomik girdiler değerlendirildiğinde çalışma sonucunda, aşı çalışmalarının ve koruyucu tedbirlerin önemi ön plana çıkmaktadır.

RESUMENLa papilomatosis bovina es una enfermedad del ganado vacuno caracterizada por la presencia de papilomas y fibropapilomas, especialmente en piel y ubres. Está originada por la infección por papilomavirus bovinos (BPV), virus desnudos de la familia Papillomaviridae.Son virus epiteliotrópicos, aunque se ha detectado su presencia en sangre, leche, orina y semen. Presentan alta diversidad viral, reconociéndose hasta la fecha 13 tipos (BPV-1 a BPV-13). Aunque originalmente se describieron en ganado vacuno, algunos genotipos (BPV-1 y BPV-2) se han asociado al desarrollo de papilomas en búfalos, cebras, jirafas y yaks. Algunos genotipos se han relacionado asimismo con el desarrollo de tumores en tracto gastrointestinal y cáncer de vejiga urinaria en ganado bovino y con sarcoides equinos. La infección por BPV se ha descrito en diferentes zonas del mundo, aunque no todos los genotipos presentan la misma prevalencia. Los trabajos más recientes muestran una elevada incidencia de infecciones múltiples.
