Sialadenitis in IQI/Jic Mice: A New Animal Model of Sjoegren's Syndrome.

Abstract: Focal infiltration of lymphocytes with parenchymal destruction was noted in both salivary and lacrimal glands of IQI/Jic mice. The sialadenitis was found in more than 80% of female mice at all ages examined. The lesion progressed after 6 months and became more prominent with age. In contrast, male mice had slight and stable salivary lesions independent of age, though the incidence increased with age. Infiltrating lymphocytes consisted of both T and B cells. The dominant lymphocytes in small foci were CD4+ cell… Show more

“…SS patients usually have lower titer of autoantibodies than those with systemic autoimmune diseases such as lupus and display a wide range of autoantibodies, with those most commonly directed against SS-A (also known as Ro) and SS-B (also known as La). Mouse models such as the NOD, MRL͞lpr, and the Id3 knockout mice have anti-SS-A and anti-SS-B antibodies (5, 7), whereas others do not (6,15). To test whether sera from r1⌬T͞r2n mice resembled the serological profile of pSS patients, we first measured antinuclear antibodies (ANAs).…”

“…Several mouse models exhibit similar pathology as pSS patients and are currently used to study the mechanism of the disease and for development of potential treatments (3)(4)(5)(6). NOD and MRL͞lpr mice are two strains that are most commonly used to model primary and secondary SS (7).…”

“…This strain is a high responder to the induction of antinucleolar autoantibody by mercuric chloride [9]. Aged female IQI/Jic mice have thymic B cells [10], and sialoadenitis similar to human Sjögren's syndrome [11]. Since female IQI/Jic mice show a higher incidence and earlier onset of SCH accompanied by prominent mast cell infiltration than other strains of mice [7], we investigated the ultrastructure of their adrenal gland.…”

“…Namely, they induce a high level of antinuclear auto-antibody following mercuric chloride treatment [21], have thymic B cells [19], and show an age-related development of Sjögren's syndrome-like sialadenitis [20]. In addition, the prevalence of allergic…”
