Serum‐derived exosomes from antigen‐fed mice prevent allergic sensitization in a model of allergic asthma

Abstract: SummaryOral tolerance is an active process that starts with sampling of luminal antigens by the intestinal epithelial cells (IEC), followed by processing and assembly with major histocompatibility complex class II and subsequently a release of tolerogenic exosomes (tolerosomes) from the IEC. We have previously shown that tolerosomes can be isolated from serum shortly after an antigen feed, and will potently transfer antigen-specific tolerance to naive recipients. Here we study the capacity of the tolerosomes t… Show more

“…Those results imply the potential roles of exosomes in immune privileges in the placenta. Tumor-specific exosomes are also found in the blood of tumor patients and their levels increase as the tumor-burden of patients increases (Allan et al, 2006;Almqvist et al, 2008;Bullerdiek and Flor, 2012;Huber et al, 2008;Karlsson et al, 2001).…”

“…MVs have been identified in the vascular circulation and can be released by many different cell types (8)(9)(10). Probably the largest fraction of MVs found in the plasma originates from endothelial and circulating blood cells (11), although the exact contribution of individual cells to circulating MVs is unknown and may be affected by different stimuli and diseases.…”

“…Mice receiving serum or isolated exosomes from ovalbumin-fed mice showed tolerance to ovalbumin exposure, because these mice had a reduced concentration of airway eosinophils, lower serum levels of total IgE and ovalbumin-specific IgE, and an increased concentration of activated T cells with a regulatory phenotypethan control animals. Thus, exosomes have the ability to prevent allergic sensitization (Almqvist et al, 2008).…”
