Some members of the gamma herpesvirus genus Macavirus are maintained in nature as subclinical infections in well-adapted ungulate hosts. Transmission of these viruses to poorly adapted hosts, such as American bison and cattle, can result in the frequently fatal disease malignant catarrhal fever (MCF). Based on phylogenetic analysis, the MCF viruses (MCFV) cluster into two subgroups corresponding to the reservoir hosts’ subfamilies: Alcelaphinae/Hippotraginae and Caprinae. Antibody cross-reactivity among MCFVs has been demonstrated using techniques such as enzyme linked immunosorbent and immunofluorescence assays. However, minimal information is available as to whether virus neutralizing antibodies generated against one MCFV cross react with other members of the genus. This study tested the neutralizing activity of serum and plasma from select MCFV-infected reservoir hosts against alcelaphine herpesvirus 1 (AlHV-1) and ovine herpesvirus 2 (OvHV-2). Neutralizing antibody activity against AlHV-1 was detected in samples from infected hosts in the Alcelaphinae and Hippotraginae subfamilies, but not from hosts in the Caprinae subfamily. OvHV-2 neutralizing activity was demonstrated in samples from goats (Caprinae) but not from wildebeest (Alcelaphinae). These results show that neutralizing antibody cross reactivity is present to MCFVs within a virus subgroup but not between subgroups. This information is important for diagnosing infection with MCFVs and in the development of vaccines against MCF.

Ovine herpesvirus-2 is a member of the gammaherpesviruses of the herpeseviridae, which is the etiologic agent of malignant catarrhal fever (MCF), a significant fatal disease of cattle. MCF disease was diagnosed in native Iraqi cattle of Al-Qadisiyah governorate of Iraq, during the period from April 2014 and August 2016. Twenty-three blood samples were collected from clinically suspected cattle. The presence of the virus in samples was ascertained based on clinical pictures, postmortem examination and molecular assays. Pansystemic involvement included respiratory, digestive, urinary, nervous systems and ocular lesions were described. A molecular analysis based on a tegument protein gene by specific semi-nested-PCR, DNA sequence and multiple alignments of all PCR products confirmed the Ovine herpesvirus-2 (OHV-2) infection, and revealed a single and double nucleotide deletion, insertion and substitutions. Some of these mutations were non-silent, resulting in changes at the predicted amino acids level into viral tegument protein. The phylogenetic analysis showed the disease was caused by two genovariants of OHV-2 including at one cluster and were related to other sequences from others countries was analyzed. MCF is sporadically occurring in cattle in Iraq, the head and eye form is more pronounced form. It has been concluded that study is provides valuable information about the genetic variation among the OHV-2 genotypes in Iraqi cattle. Based on sequence and phylogenetic analysis of tegument protein gene, this paper elucidated genetic relationship between identified Iraqi OHV-2 with other strains detected in other geographical regions. These results provide new information on the epidemiological and genetically of OHV-2 in Iraq.

ABSTRACT.-Eloi R.S.A., Marçola T.G., Paludo G.R., Araújo R.R., Colodel E.M., Lima E. Malignant catarrhal fever (MCF) is a disease caused by bovine infection with ovine herpesvirus type 2 (OvHV-2) and responsible for economic losses in different Brazilian regions. This paper describes the molecular detection of OvHV-2 by nested-PCR (nPCR) in nasal secretion/exfoliation samples and blood cell fraction (BCF) of sheep from 8 properties in the Federal District. Among the 188 nasal samples, 88 (41.5%) were positive to OvHV-2. Pregnant ewe presented no differences at the infection rate in comparison with parous females. Newly calved sheep showed higher OvHV-2 infection rate than female over 60 days of calving. Nasal samples allowed the detection of infected animals by nPCR with efficiency about twice than that in the blood cell fraction samples. In Brazil, epidemiological information about OvHV-2 infection in sheep flocks and factors involved in emergence of FCM outbreaks in cattle are still scarce. This study may provide support for elucidating some characteristics of the disease and for further epidemiological studies in the Federal District and other Brazilian States.
