Separation and purification of four chromones from radix saposhnikoviae by high‐speed counter‐current chromatography

Abstract: A preparative high-speed counter-current chromatography (HSCCC) method for the isolation and purification of 1''-O-glucosylcimifugin (1), 4'-O-beta-d-glucosyl-5-O-methylvisamminol (2), cimifugin (3) and 3'-O-glucosylhamaudol (4) from the Chinese medicinal herb radix saposhnikoviae has been successfully developed. A sample of 300 mg of crude extract was separated using ethyl acetate:n-butanol:1% aqueous acetic acid (1:4:5, v/v) as the two-phase solvent system and yielded 102.4 mg of 1 and 81.6 mg of 2. During t… Show more

“…In the published papers, many traditional extraction methods, such as ultrasonic-assisted extraction (UAE) [9], high-speed countercurrent chromatography [10], Soxhlet extraction (SE) [11] and heat-reflux extraction (HRE) have been adopted to extract chromones from S. divaricata. [12].…”

“…The major advantage of HSCCC is that it overcomes the irreversible adsorptive loss of samples onto the solid support matrix often employed in conventional methods. It also has the unique features of high recovery, high efficiency and ease of scale‐up (Liu et al , ). This method has been applied successfully to the analysis and separation of several natural products (Tong et al , ; Liu et al , ; Urbain et al , ; Destandau et al , ; Kim and Ha, ).…”

This method has been applied successfully to the analysis and separation of several natural products (Tong et al , ; Liu et al , ; Urbain et al , ; Destandau et al , ; Kim and Ha, ). There has been a report describing xanthones separation using HSCCC (Zhang and Shan, ).…”

“…Various activities of the aqueous extract of RSD have been suggested by pharmacological studies, such as suppression of adjuvant arthritis as well as in vitro anti‐convulsion, anticancer and anti‐inflammatory (Du et al , ; Du et al , ; Li et al , ; Li et al , ). Chromones, coumarins, polyacetylenes and some other compounds were isolated from RSD (Li et al , ; Liu et al , ). Because of its ‘dispelling wind, cold and dampness’ function, it is widely applied for headache, arthralgia and colitis (Dai et al , ).…”
