Separating Stem Cells by Flow Cytometry: Reducing Variability for Solid Tissues

Abstract: Summary Until there are valid identifiers that visualize stem cells in vivo, we rely upon flow cytometry to enrich for subpopulations with stem cell function. However, data reporting styles for flow cytometric analyses are typically inconsistent, creating challenges in comparing results across publications. In our view, clear reporting guidelines could improve reproducibility of stem cell analyses in solid tissues.

“…The isolation of adult stem cells in general-and MaSCs in particular-using multichannel flow cytometry is subject to variability from limited antibody specificity and technical variance (Alexander et al 2009). In addition, the distribution of mammary tissue cells, based on cell surface marker expression, changes with the developmental phase (see Fig.…”

“…tumors were analyzed on different days. In compliance with Alexander et al 49 single color controls (Lin-Cy5, CD24-FItC, CD49f-pe or pI) were included in each experiment and the compensation was done by the software. (B) Unsupervised hierarchical cluster analysis of gene expression data from tumors outgrown after sorting.…”

“…Smalley gave a talk on the pros and cons of FACS and in situ lineage tracing as approaches to the analysis of mammary epithelial subpopulations (Table 1) [4]. The importance of caution in data interpretation and being able to make ones experiments cross-comparable with results from other groups was emphasised.…”
